重组体pEF-HCV诱发小鼠体液免疫反应的研究

Humoral Immune Response to Recombinant Protein in Mice Inoculated with pEF-HCV Recombinant

  • 摘要: 目的 探讨基因疫苗在HCV感染中的预防和治疗价值.方法 将HCVC区基因插入pEF质粒EF1α启动子下游,构建pEFHCVC重组质粒,然后经肌肉注射及皮下注射免疫Balb/c小鼠.结果 在接种后3周内在肌注组内检测到抗HCV,但是抗体滴度较低,在加强免疫后7天血清抗HCV明显增高,而其它几组均未检出抗HCV.结论 构建的HCVDNA重组体可诱发Balb/c上鼠产生体液免疫应答.

     

    Abstract: Objective Study the protective and therapeutic value of DNA vaccine in HCV infection.Methods The recombinant plasmid PEF-HCV was constructed by inserting the highly conserved HCV-core gene fragment into the eukaryotic expression vector PEF under the control of a EF-1Apromoter,and then was injected into the quadriceps muscles or inoculated subcutaneously into right and left flank of Balb/c mice.Results Within 3 weeks antibody to HCV core was detected in serums of mice injected recombinant pEF-HCV core,but the titers were low and the serum antibody level rised significantly in 7 days after the booster inject ion,no specific antibody was detected in other groups.Conclusion The results suggested that HCV core DNA-based immunization can induce HCV specific antibody,but the humoral immune response was weak at early stage.
