曲酸对Vero及CHO-K1细胞毒性作用

Cyotoxic effects of kojic acid on cell lines

  • 摘要: 目的探讨曲酸对传代细胞的毒性效应。方法用不同浓度曲酸染毒Vero及CHO-K1传代细胞株,采用噻唑蓝MTT显色法于不同染毒时间终点观察细胞毒性变化。结果细胞与受试物接触24 h之内,曲酸浓度<750μg/mL,Vero细胞株对曲酸反应与阴性对照无明显差异,但随着剂量加大,毒性反应逐渐增强;随着作用时间延长,曲酸对细胞的毒性不断加大,细胞培养48 h,曲酸半数抑制效应(IE50)为2 000μg/mL;细胞培养>72 h,曲酸IE50≤500μg/mL;改变起始染毒方式,对细胞毒性效应无明显影响;CHO-K1细胞株经>1 000μg/mL曲酸染毒后,培养24 h,出现轻微抑制现象;培养48h,细胞毒性作用明显,培养72,96 h毒性反应最强烈。结论曲酸对受试细胞株具有明显毒性作用,且有明显剂量-效应和时间-效应关系。

     

    Abstract: ObjectiveTo probe the cytotoxicity of kojic acid to established cell lines.MethodsTwo cell lines,Vero cell and CHO-K1 cell,were treated with kojic acid for different time and then methyl-thiazolyl-tetrazolium assay was performed to evaluate the survival of the cells.ResultsCytotoxic effect for Vero cell with the exposure of kojic acid lower than 750μg/mL for 24 hours incubation was not observed compared with the negative control,but the dose-dependent and time-dependent response was observed after the exposure more than 24 hours.When cells were treated with kojic acid for 48 hours,50% of inhibition effect(IE50)of cell viability appeared at 2 000μg/mL,followed by IE50≤500μg/mL for more than 72 hours treatment.Cultured cells incubated with kojic acid simultaneously for 24 hours resulted in no difference in toxicity.There was a light inhibition in CHO-K1 cells exposed to kojic acid of more than 1 000μg/mL for 24 hours.With elongation of the cell incubation with kojic acid for 48,72 and 96 hours,obvious cytotoxicity to the cells was observed.ConclusionKojic acid has toxic effect on Vero and CHO-KI cell lines with significant dose-and time-response manner.
