桑叶黄酮对胰岛素抵抗大鼠氧化应激影响

Effect of mulberry flavonoids on oxidatie stress among diabetic rats with insulin resistance

  • 摘要: 目的 观察桑叶黄酮对2型糖尿病胰岛素抵抗大鼠氧化应激作用,并探讨其改善胰岛素抵抗的分子机制。方法 高脂饮食饲养成年雄性SD大鼠8周诱导大鼠胰岛素抵抗模型,随机分为糖尿病模型组(MOD组)和桑叶黄酮治疗组(MLF组),对照组为正常SD大鼠(NOR组);测量各组大鼠空腹血糖(FPG)、胰岛素(FINS)及血清丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(IRI)。结果 与NOR组比较,MOD组MDA升高,为(8.73±0.85)nmol/L,SOD、GSH-Px活性均降低,分别为(226.45±17.54)、(529.03±69.56)U/mL;与MOD组比较,MLF组能降低病鼠FINS和IR I,分别为(25.83±2.1)mU/L和(8.86±0.96),提高ISI,为(5.08±0.09),降低FPG和MDA含量,分别为(7.95±1.85)mmol/L和(6.82±0.79)nmol/mL,增强SOD活性,为(297.85±22.16)U/mL。结论 桑叶黄酮能改善2型糖尿病胰岛素抵抗大鼠高胰岛素血症,降低血糖,提高胰岛素敏感性,降低氧化应激水平,具有抗氧化能力。

     

    Abstract: Objective To observe the effect of mulberry flavone on oxidative stress in type 2 diabetes rats with insulin resistance and to explore molecular mechanism of mulberry flavone against insulin resistance.Methods The adult male SD rats were fed with high-fat diet for 8 weeks,and then randomly divided into model group with type 2 diabetes(MOD group),mulberry leaves flavone treatment group(MLF),and normal control group(NOR group).We observed levels of fasting plasma glucose(FPG),fasting insulin(FINS),serum malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)activity,insulin sensitivity index(ISI),and insulin resistance index(IRI).Results Compared with the normal group,serum MDA(8.73±0.85 nmol/L)was significantly increased;SOD(226.45±17.54 U/ml)and GSH-Px(529.03±69.56 U/ml)activity were significantly lower in model group.Compared with model group,mulberry flavone could significantly reduce FINS(25.83±2.1 mU/L)and IRI(8.86±0.96),increase ISI(5.08±0.09),lower FPG(7.95±1.85 mmol/L),and increase SOD(297.85±22.16 U/ml)activity,decrease MDA(6.82±0.79 nmol/ml) content.Conclusion Mulberry flavone can improve insulin resistance in type 2 diabetic rats with hyperinsulinemia,lower plasma glucose,improve insulin sensitivity,and resist oxidative stress.
