铅对仔鼠学习记忆及海马中IL-1β表达影响

Lead(Pb2+) impairs ability of learning and memory and affects expression of interleukin-1β(IL-1β) in hippocampus in mice

  • 摘要: 目的 探讨母体铅染毒对其仔鼠学习记忆及海马中白细胞介素-1β(IL-1β)mRNA和蛋白表达量的影响,为进一步揭示铅的神经毒性机制提供科学依据。方法 采用自由饮水的方式于母鼠孕哺期染毒,染毒剂量为0.3、1.0、3.0 g/L,每组10只;仔鼠出生后7、14、21 d分别测其血铅,海马铅浓度;仔鼠21d时,测定其学习记忆能力的变化和海马组织中IL-1βmRNA和蛋白表达。结果 各剂量染铅组血铅、海马铅含量与对照组比较明显升高(P<0.05);迷宫实验中,与对照组比较,1.0和3.0 g/L组差异均有统计学意义(P<0.05);逆转录-聚合酶链反应(RT-PCR),ELISA实验中,在对照组0.3、1.0、3.0 g/L组仔鼠海马组织中,IL-1βmRNA的相对表达量分别为(0.073±0.016)、(0.572±0.287)、(1.090±0.521)、(1.312±0.837);IL-1β蛋白表达分别为(0.708±2.260)、(8.958±2.257)、(19.375±1.644)、(25.125±4.748);与对照组比较,染毒组海马IL-1βmRNA和蛋白表达量明显升高(P<0.05)。结论 铅可能通过诱导海马中IL-1β高表达,影响学习记忆,从而造成神经系统的损害。

     

    Abstract: Objective To explore the effect of lead on learning,memory and the expression of IL-1βmRNA and protein in rats for the study of neurotoxic mechanism of lead.Methods Pregnant mice(10 for each group)were exposed to low(0.3 g/L),moderate(1.0 g/L),and high-level(3.0 g/L)of lead acetate in deionized drinking water from the first day of gestation to 21th postnatal day(PND).The lead concentration in blood and hippocampus were determined at 7th,14th, 21th day of neonatal mice pups.The ability of learning and memory and the expression of IL-1βmRNA and protein in hippocampus of different groups at 21th day were measured.Results The lead concentration in lead exposure group was higher than that of in control group(P<0.05).In water Morris maze test,there was statistical significance in moderate and high lead exposure group(P<0.05).The relative expression level of IL-1βmRNA in control group,low,moderate,high lead exposure group were 0.073±0.016,0.572±0.287,1.090±0.521,and 1.312±0.837 and the expression of IL-1β protein was 0.708±2.260,8.958±2.257,19.375±1.644,and 25.125±4.748,respectively.The expressions of IL-1β mRNA and protein were higher in lead exposed groups than that of in the control group(P<0.05).Conclusion Lead may affect learning and memory and cause nervous system damage by inducing IL-1β expression in hippocampus in mice.
