柴胡皂苷D对SGC-7901细胞生长及迁移抑制作用

Inhibitory effect of saikosaponin D on growth and migration of gastric cancer cell line SGC-7901

      目的  探讨柴胡皂苷D对胃癌SGC-7901细胞生长、迁移抑制作用及机制。
      方法  实验设对照组、低、中、高剂量柴胡皂苷组(5、10、20 μmol/L柴胡皂苷D);采用四甲基偶氮唑蓝(MTT)法检测各组细胞24、48和72 h细胞增殖情况;采用原位末端转移酶标记法(TUNEL)检测各组细胞凋亡; Transwell检测各组细胞迁移能力;实时定量荧光PCR技术(real time PCR)检测各组细胞中norrin及livin表达;蛋白印迹法检测各组细胞norrin、livin、p57和cyclin E表达。
      结果  MTT结果显示,与对照组比较,低、中、高剂量柴胡皂苷组SGC-7901细胞增殖受到明显抑制,呈剂量效应关系(P < 0.05);Transwell结果显示,与对照组(190 ± 7)个/视野比较,低、中、高剂量柴胡皂苷组细胞迁移数量分别为(114 ± 9)、(87 ± 6)、(38 ± 4)个/视野均减少(均P < 0.05);与对照组(4.94 ± 1.14)比较,低、中、高剂量柴胡皂苷组SGC-7901细胞凋亡指数分别为(8.46 ± 1.18)、(11.76 ± 2.85)、(34.34 ± 7.74)均增加(均P < 0.05);与对照组比较,中、高剂量柴胡皂苷组SGC-7901细胞中norrin、livin mRNA及蛋白表达均减少(均P < 0.05),各剂量组SGC-7901细胞中cyclin E 蛋白表达均明显减少,p57表达明显增加,且呈剂量效应关系(均P < 0.05)。
      结论  柴胡皂苷D对胃癌细胞生长、迁移具有抑制作用,其机制可能与柴胡皂苷D减少norrin、livin表达,延长细胞周期、促进细胞凋亡有关。

     

    Abstract:
      Objective  To investigate inhibitory effect and machanism of saikosaponin D (SSd) on the growth and migration of gastric cancer SGC-7901 cells.
      Methods  The SGC-7901 cells were assigned into a control group and low-, moderate-, and high-dose SSd (5, 10, and 20 μmol/L) groups. The cell proliferation was detected with methyl thiazolyl tetrazolium (MTT) assay at 24, 48 and 72 hours of cell culture. The cell apoptosis was measured with terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling (TUNEL). The migration ability of the cells was determined with Transwell. The expression of norrin and livin mRNA were determined with real-time PCR. The cellular expression of norrin, livin, p57, and cyclin E were detected with Western blot.
      Results  The results of MTT assay revealed that the proliferation of the cells treated with SSd was significantly inhibited in a dose-response manner (P < 0.05). Transwell results showed that the number of migration cells of low-, moderate-, and high-dose SSd groups (114 ± 9, 87 ± 6, and 38 ± 4 per field of view) were significantly lower than that of the control group (190 ± 7 per field of view) (P < 0.05 for all); whereas, the cellular apoptosis of the different SSd dose groups (apoptosis index AI: 8.46 ± 1.18, 11.76 ± 2.85, and 34.34 ± 7.74) were significantly increased compared to that of the control group (AI: 4.94 ± 1.14) (all P < 0.05). Compared with the control group, the expressions of norrin and livin mRNA and protein in the cells of moderate- and high-dose SSd groups decreased significantly compared with those of the control group (P < 0.05 for all). The expression of cyclin E protein in cells treated with SSd was significantly decreased but the expression of p57 was significantly increased, with a dose-effect relationship (P < 0.05 for all).
      Conclusion  Saikosaponin D can inhibit the growth and migration of gastric cancer cells by promoting cell apoptosis and affecting cell cycle.
