金银花菌子实体及发酵菌丝对小鼠免疫调节作用比较

Effects of sporocarp and fermentative hyphae of Phylloporia ribis on immunoregulation in mice: a comparative study

      目的  比较金银花菌子实体及发酵菌丝对ICR正常小鼠的免疫调节作用。
      方法  分别以1.25、2.50、5.00 g/kg金银花菌发酵菌丝粉和1.25、2.50、5.00 mL/kg金银花菌子实体提取液经口给予小鼠,连续30 d,通过足趾增厚法、小鼠脾淋巴细胞转化实验检测小鼠细胞免疫调节能力,通过溶血素实验、抗体生成细胞实验检测小鼠体液免疫调节能力,通过巨噬细胞吞噬实验、自然杀伤细胞(NK)活力实验测定小鼠非特异性免疫调节能力。测定脾指数和胸腺指数。
      结果  与对照组比较,金银花菌子实体各剂量组及发酵菌丝中、高剂量组小鼠的足趾肿胀度明显增加(P < 0.01),子实体及发酵菌丝高剂量组小鼠淋巴细胞转化能力明显提高(P < 0.05);与对照组比较,子实体及发酵菌丝中、高剂量组小鼠抗体生成细胞数量明显增加,子实体与发酵菌丝高剂量组小鼠碳廓清能力及子实体高剂量组与发酵菌丝中、高剂量组小鼠NK细胞活性均明显增加 (均P < 0.01);与对照组比较,子实体中、高剂量组及发酵菌丝高剂量组小鼠腹腔巨噬细胞吞噬百分率与吞噬指数明显升高。
      结论  一定剂量的金银花菌子实体及发酵菌丝均能明显增强小鼠细胞免疫、体液免疫、非特异性免疫能力;子实体在体液免疫上优于发酵菌丝;发酵菌丝在非特异性免疫上优于子实体。

     

    Abstract:
      Objective  To compare immunomodulatory effects of sporocarp and fermentative hyphae of Phylloporia ribis in normal ICR mice.
      Methods  The experimental mice were orally administrated with low-, moderate-, and high-dose suspension (1.25, 2.50, and 5.00 g/kg·body weight) of fermentative hyphae and sporocarp extracted from Phylloporia ribis, respectively for 30 days consecutively. The cellular immunoregulation of the mice was verified with delay-type hypersensitivity (DTH) test and proliferation and transformation of spleen lymphocyte. The humoral immunoregulation was assessed with antibody-producing cell and serum hemolysin level test. Nonspecific immune regulation was monitored with carbon clearance, macrophage phagocytosis test and activity of natural killer (NK) cells. Spleen index and thymus index of the mice were also determined.
      Results  Significantly increased degree of swelling of toes was observed in all sporocarp groups and moderate- and high-dose fermentative hyphae groups compared to that in the control group (all P < 0.01). Lymphocyte transformation ability was obviously increased in the mice of sporocarp groups and high-dose fermentative hyphae group (P < 0.05 for all). Compared to that of control group, the number of antibody-producing cell of all sporocarp and moderate- and high-dose fermentative hyphae groups increased significantly (all P < 0.01). The carbon clearance capability of sporocarp groups and high-dose fermentative hyphae group and NK cell activity of sporocarp and moderate- and high-dose fermentative hyphae groups were promoted significantly (all P < 0.01). The percentage and index of peritoneal macrophage phagocytosis of moderate- and high-dose sporocarp groups and high-dose fermentative hyphae group were elevated remarkably.
      Conclusion  Sporocarp and fermentative hyphae of Phylloporia ribis could promote, at certain dosage, cellular and humoral immunity and nonspecific immunocompetence in mice; the humoral immunomodulatory effect of sporocarp is stronger than that of fermentative hyphae, while the nonspecific immunomodulatory effect of fermentative hyphae is stronger than that of sporocarp.
