咳必停镇咳祛痰平喘作用

Antitussive, expectorant and anti-asthmatic effects of Kebiting: a pharmacodynamics study

      目的  观察咳必停(KBT)是否具有镇咳、祛痰、平喘作用。
      方法  应用正交试验设计确定咳必停中白芥子、紫苏子、莱菔子的最佳组合剂量。咳必停高、中、低剂量组与阳性对照组(桂龙咳喘宁)及空白组进行对照。用小白鼠酚红法观察咳必停的祛痰作用;小白鼠氨水引咳法、豚鼠枸橼酸引咳法观察咳必停的镇咳作用;豚鼠组胺和乙酰胆碱引喘法观察咳必停的平喘作用。
      结果  咳必停最佳剂量组合为因素A1水平、因素B1水平、因素C2水平。咳必停各剂量组的酚红排出浓度均高于空白对照组(P < 0.01),中、高剂量组明显高于阳性对照组;咳必停各剂量组小鼠的咳嗽潜伏期均明显长于空白对照组(P < 0.01),咳嗽次数明显少于空白对照组(P < 0.01),中、高剂量组镇咳作用明显高于阳性对照组(P < 0.01);咳必停低、高剂量组豚鼠的咳嗽次数与空白对照组相比差异显著(P < 0.01),中剂量组作用稍弱(P < 0.05);咳必停各剂量组的引喘潜伏期与空白对照组相比差异显著(P < 0.01),中剂量组潜伏期明显长于阳性对照组(P < 0.01)。
      结论  咳必停具有明显的镇咳、祛痰、平喘作用。

     

    Abstract:
      Objective  To evaluate whether Kebiting (KBT, a compound preparation made of three Chinese medicinal herbs) has antitussive, expectorant and anti-asthmatic effect.
      Methods  Mice and guinea pigs were used to conducted the animal study. Orthogonal experimental design was used to determine the optimal combination components of Brassica alba, Perilla frutescens, and Semen raphani of the KBT. The animals were divided into high, moderate, and low doses of KBT groups, a positive control (treated with Guilongkechuanning, a Chinese patent medicine for the treatment of cough, phlegm, and asthma) and a blank control group. Animal model of cough was established with administration of ammonia and citrate in the mice and guinea pigs and guinea pig asthma model was established with histamine and acetylcholine. Expectorant effect of KBT in mice was observed with phenol red method.
      Results  The optimal combination of three components of KBT was constructed. The excretion of phenolsulfonphthalein in the mice of all KBT groups was significantly higher than that of the blank control group (P < 0.01); the excretion of phenolsulfonphthalein in the mice with high dose KBT was significantly higher than that of the positive control group (P < 0.01). The latent period of cough was prolonged and the frequency of cough was decreased significantly in all the mice of KBT groups compared with those of the blank control group (both P < 0.01), with a significantly stronger antitussive effect for moderate and high dose of KBT in comparison with positive control treatment. Significantly decreased cough frequency was observed in KBT treated guinea pigs, with significant differences in high and low dose groups (P < 0.01) and in moderate dose group (P < 0.05) both compare to that of the blank control group; the asthma latent period of all KBT treated guinea pigs was also significantly longer than that of the blank control group (P < 0.01).
      Conclusion  KBT has significant effects of antitussive, expectorant, and anti-asthma in mice and guinea pigs.
