章鱼蛋白钙对小鼠补钙功效评价

Effect of octopus protein calcium on calcium supplement in mice

    目的 研究章鱼蛋白钙(OP-Ca)对小鼠的补钙功效。
    方法 将60只ICR小鼠随机分为5组,分别为正常对照组、低钙组、碳酸钙组、OP-Ca低、高剂量组。除正常对照组外,其余4组喂饲低钙饲料。低、高剂量OP-Ca组分别灌胃给予OP-Ca 39.9、119.7 mg/kg,碳酸钙组灌胃给予碳酸钙119.7 mg/kg,正常对照组和低钙组灌胃给予等体积去离子水,每日1次,连续4周,并进行小鼠钙代谢实验,收集小鼠的粪便和尿液,测定粪钙和尿钙含量,计算钙的表观吸收率和储留率。4周后处死小鼠,眼球取血,分离血清,取小鼠双侧股骨,测定血清Ca、股骨钙含量等指标。
    结果 低钙组血清钙、股骨钙含量分别为(1.98 ± 0.10)mmol/L、(217.24 ± 13.30)mg/g,明显低于低、高剂量OP-Ca组(P < 0.05);高剂量OP-Ca组钙表观吸收率、储留率、血清Ca、股骨钙分别为(76.30 ± 2.44)%、(75.29 ± 2.11)%、(2.23 ± 0.10)mmol/L、(249.12 ± 12.39)mg/g,均显著高于碳酸钙组(P < 0.05)。
    结论 OP-Ca具有较高的生物利用率,可显著提高小鼠的骨钙含量,改善其钙营养状况,补钙效果优于碳酸钙。

     

    Abstract:
    Objective To study the calcium supplementation effect of octopus protein calcium (OP-Ca) in mice.
    Methods Sixty ICR mice were randomly divided into 5 groups: normal control (with deionized water), low calcium (with deionized water), calcium carbonate (at dosage of 119.7 mg/kg), and low- and high-dose OP-Ca groups (with OP-Ca of 39.9 and 119.7 mg/kg) and the treatments were carried out by gavage once a day for 4 weeks. All the mice were supplied with low-calcium feed except for normal control mice. Calcium content in feces and urine of the mice were measured to calculate calcium absorption and retention rate for assessing calcium metabolism of the mice. Eyeball blood and the bilateral femurs samples of the mice were collected by the end of intragastric administration to detect serum and bone calcium.
    Results The serum calcium (1.98 ± 0.10 mmol/L) and femur calcium content (217.24 ± 13.30 mg/g) in the mice of low calcium group were significantly lower than those in the mice of low- and high-dose OP-Ca groups (both P < 0.05). Compared with calcium carbonate group, the mice of high-dose OP-Ca group had significantly increased apparent calcium absorption rate (76.30 ± 2.44 %), calcium retention rate (75.29 ± 2.11%), serum calcium (2.23 ± 0.10 mmol/L) and femur calcium content (249.12 ± 12.39 mg/g) (P < 0.05 for all).
    Conclusion With a high bioavailability and a better supplement effect than calcium carbonate, octopus protein calcium could significantly increase bone calcium content and improve calcium nutritional status in mice.
