少数民族6~16岁在校学生铁营养状况关联基因多态性地域特征分析

Regional feature of iron nutrition-related gene polymorphisms among 6 – 16 years old minority students

      目的  研究中国少数民族在校学生人群营养素缺乏风险的关联SNP位点地域特征。
      方法  搜集以往研究中报道的21个SNP位点,采用2016年广西、内蒙和新疆地区384名贫困农村学生的凝固血细胞,磁珠法提取DNA,Sequenom MassArray飞行质谱检测系统进行基因分型检测,采用SPSS 18.0软件对数据结果进行分析。
      结果  10个SNP位点分型结果在地域分布上差异有统计学意义。3号和22号染色体上各3个位点进行的连锁不平衡和单倍型分析结果显示,3号染色体上新疆地区rs3811658和rs1830084之间的r2为0.81,其他2个地区r2 > 0.8。 22号染色体上rs2111833和rs1421312之间r2分别为壮族0.30,蒙古族0.24,维吾尔族0.44,结果显示壮族和维吾尔族在校学生中2个位点间显示连锁不平衡强关联性,蒙古族在校学生位点连锁不平衡程度弱于壮族和维吾尔族。
      结论  3个少数民族在校学生铁营养状况关联基因多态性存在区域差异。

     

    Abstract:
      Objective  To explore nutrient deficiency-related gene single nucleotide polymorphism (SPN) loci and their regional distribution among Chinese minority students.
      Methods  We selected 21 SNP loci for the analysis based on studies published previously. We collected coagulated blood cell samples among 384 minority students from poverty-stricken rural areas in Guangxi Zhuang Autonomous Region, Inner Mongolia Autonomous Region, and Xinjiang Uygur Autonomous Region (Xinjiang) in 2016 and extracted DNA of the blood cells with immunomagnetic bead method. We performed genotyping with Sequenom Mass Array system and conducted data analyses with SPSS 18.0.
      Results  Significant differences in geographical distribution of SNP were observed in 10 loci of nutrient deficiency-related genes. Linkage disequilibrium (LD) and haplotype analysis resulted in the r2 value of 0.81 for the LD between rs3811658 and rs1830084 loci on chromosome 3 among Uygur students in Xingjian but the r2 values were less than 0.8 for students in the other two regions. The r2 between rs2111833 and rs1421312 on chromosome 22 was 0.30 for the Zhuang, 0.24 for the Mongolian, and 0.44 for the Uygur students, indicating a strong LD correlation between the two loci among Zhuang and Uygur students and a weaker LD correlation of the two loci among Mongolian students.
      Conclusion  The study results demonstrate regional differences in SNP of iron nutrition-related genes among Uygur, Zhuang, and Mongolian students.
