CXCL12基因在肾透明细胞癌的表达特征 及作用机制

Expression and effect mechanism of CXCL12 gene in kidney renal clear cell carcinoma: a bioinformatics study

      目的  探讨趋化因子CXCL12基因在肾透明细胞癌的表达特征及作用机制。
      方法  利用UALCAN数据库分析肾透明细胞癌中CXCL12基因及蛋白表达情况;利用TCGA数据库获取肾透明细胞癌中的CXCL12基因、BMP家族基因表达与临床预后的关系,采用Kaplan-Meier法绘制生存曲线;筛选肾透明细胞癌中CXCL12高表达组/低表达组差异表达基因并进行GO分析,获得相关信号通路。
      结果   CXCL12基因及相应蛋白在肾透明细胞癌组织中呈低表达(P < 0.0001);CXCL12基因高表达患者总体生存率和疾病特异性生存率均高于CXCL12基因低表达患者(P < 0.05);CXCL12高表达组的上调基因主要集中在骨形态发生蛋白(BMP)相关的信号通路中;进一步发现CXCL12与4个BMP家族基因(BMP6、 BMPR2、 BMP2K、 BMPR1A)呈正相关(P < 0.0001),且这4个BMP家族基因高表达的患者总体生存率均增高(P < 0.05)。
      结论   CXCL12基因在肾透明细胞癌疾病进程中发挥重要作用,与BMP家族基因密切相关。

     

    Abstract:
      Objective  To investigate the expression and effect mechanism of CXCL12 gene in kidney renal clear cell carcinoma.
      Methods   UALCAN database was used to analyze the expression of CXCL12 gene and protein in kidney renal clear cell carcinoma. TCGA database was used to obtain the expression of CXCL12 gene, BMP family gene and clinical information, and Kaplan-Meier was used to analyze the survival curve. The differentially expressed genes in the CXCL12 high/low-expression groups in kidney renal clear cell carcinoma were screened and GO analysis was performed to obtain relevant signaling pathway.
      Results  Lower expressions of CXCL12 gene and relative protein were found in the tissues of kidney renal clear cell carcinoma. The overall survival rate and disease specific survival rate in the patients with high expression of CXCL12 gene were significantly higher than those in the patients with low CXCL12 gene expression. The majority of the up-regulated genes associated with high CXCL12 gene expression were identified in BMP-related pathway. The expression of CXCL12 is positively correlated with four BMP family genes and significantly higher overall survival rate was observed in the patients with higher expressions of any of the four BMP family genes.
      Conclusion  CXCL12 gene may play an important role in the progression of kidney renal clear cell carcinoma and CXCL12 gene expression is associated with BMP family genes.
