东北地区恙虫病东方体分离株sta56基因片段的扩增及序列分析

Amplification and Sequencing of Partial Sta56 Gene of Orientia Tsutsugamushi Strains Isolated in North east Areas

  • 摘要: 为了解东北地区恙虫病东方体分离株与其参考株的同源关系,采用了PCR技术扩增东北地区恙虫病东方体分离株sta56基因片段,测定HCAa9202和HCM9501株sta56基因片段的核苷酸序列,并与已知列的参考株进行同源性比较.结果显示,东北地区恙虫病东方体分离株均可在320~332bp范围内见到特异性扩增带,但MSAa9302株和MSAa9304株在此范围内分别可见2条和3条扩增带.HCAa9202株和HCM9501株与6个已知序列的恙虫病东方体菌株相比较同源性在67.72%~83.18%间,相对与Gilliam的同源性较高.

     

    Abstract: In order to get a clear understanding of nucleotide homologous relation between isolated strains in North-east area and other reference strains of Orientia tsutsugamushi,partial sta56 gene of the isolaled strains were amplified by PCR.Amplified-fragment of isolated strains HCAa9202 and HCM 9501 were sequenced.Meanwhile,a homologous comparison was made with the reference strains of known sequence.Results showed that a specific amplified-fragmants of the Orientia T sutsugamushi strains isolated in Northeast ar ea was observed in the fields of 320~332bp,while 2~3 amplified-fr ag mant were observed seperately in the strains of MSAa9302 and MSAa9304 in the same fields.The nucleotide homolog y between the strains of HCAa9202,HCM 9501 and the 6 Orientia T sutsugamushi strains of known sequence was 67.72%~83.18%.Both isolated strains have a high nucleotide homology with Gilliam strain.
