氧化型染发剂涂敷家兔皮肤毒理实验研究

Study on Toxicity of Rabbits’ Skin Painted with Oxidizing Hair-dyes

  • 摘要: 目的 研究S牌染发剂对家兔的毒性作用。方法 健康白色家兔经皮涂敷S牌染发剂6个月(每周涂抹2次),解剖动物,取内脏组织作病理切片、免疫组化及细胞周期分析。结果 动物解剖,未发现癌肿。HE染色后病理组织切片观察肝、脾、肾、胸腺、淋巴结、心脏各组织,均未见异常。免疫组化检测发现:肝、脾、淋巴结各组织中P53、PCNA及BCL-2均呈不同程度的阳性反应。细胞周期分析发现:染发剂组家兔肝、脾、淋巴结3种组织除了二倍体细胞群外,还有1组异倍体细胞群。结论 某些氧化型染发剂对家兔可能具有潜在的致癌作用。

     

    Abstract: Objective To study the toxicity of S-brand hair dyes on rabbits.Methods S-brand hair dyes was spread on the skin of rabbits for six months(two application per week).Rabbits were dissected,Sections from liver,spleen,kidney,thymus,lymphnode and heart were detected from microscope after HE staining,and cell cycles were also analysed.Results No carcinoma was found in all of the rabbits dissected.No abnormal changes were found in all the tissues.Immunohis-tochemical study of liver,spleen and thymus node showed that nucleus positivity of P53 in all the three tissues are higher than the normal control group.Cytoplasm positivity of BCL-2 in these three tissues were also higher than the control group.Cell cycle analysis found that all the three tissues(rabbits applied with hair-dyes)have aneuploid except normal diploid.Conclusion S-brand hair-dyes didn't induce carcinoma in the rabbits,but it was a potential carcinogen.
