磺酸左氧氟沙星对细胞内HBV S区及C区HBV RNA的影响

Effect of Levofloxacin Sulphonate on HBV RNA of 3.5kb and 2.1kb in 2.2.15 Cell Line

  • 摘要: 目的 研究磺酸左氧氟沙星对2.2.15细胞内HBV S区及C区的HBV RNA的影响.方法 用荧光PCR定量法分别检测细胞内HBV DNA、HBV S区及C区的HBV RNA含量;并用固相放射免疫法测细胞上清HBsAg、HBeAg的P/N值.结果 2.2.15细胞内HBVS区及C区的HBVRNA含量分别为0.4pg/ml和0.27pg/ml,细胞上清HB-sAg及HBeAg分别为42.1和36.0.磺酸左氧氟沙星作用8天后,HBV RNA含量分别下降为0.2pg/ml和0.13pg/ml(P<0.05),对两者的抑制率分别为50%和48.1%;HBsAg及HBeAg分别下降为6.0和16.2(P<0.05),其抑制率分别为85.7%及55.6%(P<0.05),用药前2.2.15细胞内HBV DNA为2.97pg/ml,用药后降至0.44pg/ml(P<0.05),抑制率为85.2%.结论 磺酸左氧氟沙星抗HBV的作用可能是通过影响HBV 3.5kb及2.1kb RNA的稳定性及翻译表达来实现.

     

    Abstract: Objective To study the effect of levofloxacin sulphonate on HBV RNA of 3.5kb and 2.lkb in the 2.2.15 cell line.Methods HBV DNA,HBV RNA were examined by quantitative PCR;HBsAg and HBeAg were tested by radioimmunoassay.Results The quantity of HBV DNA,3.5kb HBV RNA and 2.1kb HBV RNA were 2.97pg/m1,0.40pg/ml and 0.27pg/ml respectively in the 2.2.15 cell line; 0.44pg/ml,0.20pg/ml and 0.13pg/ml respectively after treated by levofloxacin sulphonate(P<0.05 );the inhibited rate were 85.2%,50.0 0.6 and 48.1% respectively.The value of HBsAg and HBeAg were 42.1 and 36.0 respectively in the 2.2.15 cell superrant;6.0 and 16.2 respectively after treated by levofloxacin sulphonate,the inhibited rate were 85.7% and 55.6% respectively(P<0.05).Conclusion The effect of levofloxacin sulphonate against HBV infection may be through affecting the stability and translation of 3.5kb,2.1kb HBV RNA in the 2.2.15 cell line.
