HFRS病毒滇绒鼠分离株E-142全S基因序列分析

Sequences Analysis of Complete S gene of Hantavirus E-142 Strain from Yunnan E.eleusis

  • 摘要: 目的了解肾综合征出血热病毒云南地区滇绒鼠分离株E-142的分子生物学特征.方法应用PCR方法克隆了该株全S基因.结果序列测定结果表明E-142株的S片段的全基因序列共1702个核苷酸,编码429个氨基酸.四种核苷酸的比例分别为A32.79%,G25.12%,T23.02%,C19.07%,GC含量为44.19%,AT含量为55.81%.E-142与汉坦病毒S基因核苷酸和氨基酸同源性比较表明,E-142株与HTN型同源率为86.6%~90.4%,而与SEO型仅为68.3%~69.6%,E-142株病毒属于HTN型,但与HTN型其它毒株相差高达9.6%~13.4%.结论E-142株病毒是HTN型的新亚型毒株.

     

    Abstract: ObjectiveFor understanding the molecular characteristics of Hantavirus E-142 strain isolated from Yunnan E.Eleusis.MethodsThe S complete gene was amplified by RT-PCR and the purified products was cloned into Tvector.ResultsComplete S segment of E-142 was 1 702 nucleotides in length encoding 429 amino acids.It showed 86.6%-90.4% homology with HTN virus,but only 68.3%-69.6% homolgy with SEO viruses at nucleotide level.ConclusionThe results suggested that the E-142 strain was a new subtype of HTN virus.
