微囊藻毒素促肝癌过程中肝细胞PCNA、p21waf1基因表达研究

Study on Expression of PCNA and p121waf1 in Course of MC Promoting Liver Tumor

  • 摘要: 目的探讨微囊藻毒素(MC)促肝癌的分子机制.方法采用二阶段致癌理论建立中期试验动物模型,γ-GT染色检验MC的促癌作用,并以免疫组化法结合图像分析技术精确测定大鼠肝脏PCNA和p21waf1基因的表达情况.结果(1)MC在促大鼠肝癌过程中能显着增加γ-GT阳性率.(2)MC在促大鼠肝癌过程中能显着增加大鼠肝脏PCNA基因表达强度和面积.(3)MC在促大鼠肝癌过程中能显着降低大鼠肝脏p21waf1基因的表达强度和面积.结论(1)进一步证明MC促癌剂的作用.(2)初步明确,调节与细胞增殖相关的癌基因和抑癌基因表达是MC促癌过程的重要机制.

     

    Abstract: ObjectiveTo explore the molecular mechanism of MC in the course of MC promoting liver tumor.MethodsTwo-stage-medium-term theory were applied to set up the animal model,and biochemistry technique to evaluate the model,and then immunohistochemical technique and image analysis were used to study the expression of the PCNA and p21waf1 in the course of promoting tumor.Results(1)MC could enhance the positive reaction ration of γ-GT.(2)MC could increase the expression of PCNA.(3)MC could decrease the expression of p21waf1.Conclusion(1)MC can promote liver tumor strongly.(2)The expression change of PCAN and p21waf1 played an important role in the course of MC promoting liver tumor.
