  • 摘要: 目的探讨McAb-ELISA检测丝虫特异IgG4在丝虫感染监测中的应用价值.方法利用生化免疫技术,经细胞融合,建立起7株对IgG4表现为单一反应性的单克隆抗体(McAb).以此为探针,采用ELISA方法检测丝虫特异IgG4.结果检测班氏微丝蚴血症者血清的敏感性为96.3%(104/108),测得IgG4的最低限量为0.01μl/ml;检测丝虫病非流行区健康者、肠道线虫感染者、华支睾吸虫感染者和囊虫病患者等血清均呈阴性,未出现交叉反应,特异性为100%.经现场应用研究,同样获得良好的实用性,有效地反映出当地丝虫病流行与感染状况.结论该技术是监测丝虫感染的理想方法.

     

    Abstract: ObjectiveTo discuss the practical value of McAb-ELISA to detect filaria speicfic IgG 4.MethodsSeven strains of filaria-specific McAb were established through technique of cellular fusion and biochemistry-immunolo gy,and were used in filarial infection test as probes to detect filaria-specific IgG4.ResultsIts sensitiv ity was 96.3% (104/108).It detected as low as 0.01μl/ml IgG4 per milliliter serum.No cross-action were tested with serums collected from residents of non-endemic area,including those infected with intest inal nematodes,liver fluke and systicercus.Its specificity was 100%.It was also feasible and practical when applied to field study,effectively reflected the epidemic situation of local filarial infection.ConclusionThis was an ideal method to be used in filarial infection surveillance.
