凹顶藻提取物的抗氧化作用

Effects of Laurencia extract on antioxidant activities in mice

  • 摘要:
      目的   观察凹顶藻提取物(LET)对DNA损伤和机体抗氧化能力的影响.
      方法   对小鼠补充不同剂量凹顶藻提取物(LET), 采用标准化方法制备LET, 以反相高效液相色谱法测定总萜含量, 并采用Horn法检测其半数致死量(LD50).以体内实验进行提取物的抗氧化活性评价, 采用单细胞凝胶电泳法测定淋巴细胞的DNA损伤, 以试剂盒测定血浆中超氧化物歧化酶(SOD)、丙二醛(MDA)含量, 应用SPSS11.5软件进行数据处理.
      结果   LET中的总萜含量为63.29%.其LD50 > 3160mg/kg, 属于低毒.LET中剂量组的血淋巴细胞DNA损伤与高剂量组和空白对照组比较差异有统计学意义(P < 0.05).LET各剂量组小鼠血中MDA的含量与空白对照组比较均降低, 差异有统计学意义(P < 0.05).
      结论   LET含有丰富的天然萜类化合物, 且使用安全; 中剂量LET对H2O2诱导的血淋巴细胞DNA氧化损伤有明显的保护作用; LET可降低血中MDA含量, 有一定的抗脂质过氧化作用.

     

    Abstract:
      Objective   The antiox idant activities of various dosages of Laurencia extract(LET)were investigated in mice.
      Methods   The total content of ter penoids in the LETwas detected by High Performance Liquid Chro matogr aphy (HPLC)and its mediallethaldose(LD50)was estimated by Horn assay.The antiox idant activit ies of LETwere detected in mice.The oxidative DNA damage of peripher al lymphocytes induced by H2O2 was analyzed by SCGE.The concentrations of superoxide dismutase(SOD), malonyl dialehyde(MDA)in plasma were detected withe test kit.Data were analzyed by SPSS software.
      Results   The concentration of total ter penodis in the LET was proved to be 63.29%.The LETshowed lo wtoxicity with its LD50 more than 3 160 mg/kg.There was a significant decreasing tendency of the oxidative DNA damage on peripheral lymphocytes in the 50 mg/kg supplement group induced by 10 mol/LH2O2.The lev els of MDA in the supplement groups were less than those of the control group.
      Conclusion   The LET rich in ter penoids is safety to be taken o rally.The 50 mg/kg LET can protect the peripherally mphocytes from oxidative DNA damage induced by H2O2.The LEThas anti lipid peroxidation activity.
