人类疱疹病毒6B型纯化鉴定方法比较和优化

Comparison and optimization of identification methods after purification of Human herpesvirus-6B

  • 摘要:
      目的   通过对人类疱疹病毒(HHV)-6B纯化后鉴定手段的比较, 筛选或优化出简单、快速、准确特异的方法.
      方法   采用蔗糖密度梯度离心法进行病毒纯化; 通过PCR法、斑点杂交法、蛋白免疫印迹法对纯化样品中HHV-6B进行鉴定, 以确定收集阳性样品的范围.
      结果   3种方法都可鉴定出阳性病毒体, 但是在样品的阳性收集范围、鉴定所需时间以及成本上表现差异.
      结论   纯化HHV-6B后的初步鉴定, 最好采用PCR法和斑点杂交法.

     

    Abstract:
      Objective   To select or optimize simple, fast and special means by compare of identification metheods after purification of Human herpesvirus-6B.
      Methods   The metheod of sucrose gradient centrifugation was used to purify Human herpesvirus-6B(HHV-6B); and in order to make sure the range of positive samples collection, the viron was detected by the metheods of PCR, Dot blotting, Western blotting.
      Results   Above metheods which all could detect the positive virus from samples, there were significant difference between the range of positive samples collection, time of identification and cost and so on.
      Conclusion   It is better to use the PCRand Dot blotting to primary identify the virus after HHV-6Bpurification.
