Abstract:
Objective To explore the effectof fish oil and transmembrane TNFA eukaryotic expression vector on breastcancerx enograft growth in nude mice.
Methods MCF-7 cells were transplanted subcutareously to BALB/c nude mice to produce tumor.The nude mice with x enograftwere randomly divided intofour groups:control group, fish oil treated group, TNFA transfacted group and fish oil+TNFA transfacted group.The xenog rafttumor growth in mice was observed.The cell cycle was measured by using flowcy to meter.The expression of transmembrane TNFAmRNA was deter mined by RT-PCR.
Results Fish oil significantly inhibited the tumor growth in vivo.The cell cycle analysis showed thatfish oil blocked the tumor cells atthe G1/G0 phage so as to reduce the ratio of Sphage.These chang es were more obviously in fish oil+TNFA transfacted group than those of fish oil treated group.The expression of transmembr ane TNFA mRNA was increased in fish oil +TNFA transfacted group.
Conclusion Fish oil could induce expression of transmembrane TNFA in xenog rafttumor transfacted with pPPRE-tk-tmTNFA, and this could promote the anti-tumor activity of fish oil in vivo.
