燃煤型砷中毒患者金属硫蛋白基因表达

Study on gene expression of metallothionein in patients suffered from coal-burnt arsenism

      目的   比较燃煤型砷中毒患者与正常人群金属硫蛋白(MT)表达, 研究金属硫蛋白基因在燃煤型砷中毒患者中的表达及其意义, 阐明燃煤型砷中毒分子机制。
      方法   燃煤型砷中毒患者与正常人群各50例, 采用Trizol-酚-氯仿一步法提取总RNA。紫外分光光度法测定RNA的量, A260/A280鉴定RNA纯度。采用实时荧光RT-PCR技术测定燃煤型砷中毒MT mRNA的表达, 以-βactin作为实时荧光RT-PCR的质控, 当P < 0.05时认为差异有统计学意义。
      结果   正常组MT mRNA的相对表达水平为6.82±7.80, 砷中毒组MT mRNA的相对表达水平为2.02±2.82。燃煤型砷中毒患者MTmRNA表达降低, 约为正常对照的30%, 二者间差异有统计学意义(P < 0.01)。
      结论   金属硫蛋白在燃煤型砷中毒代谢中有重要意义, 起重要的调控作用。MT mRNA表达降低可能是砷中毒机制之一。

     

    Abstract:
      Objective   To elucidate the molecular mechanism of arsenism caused by burning coal, Gene expriession of Metal lothionein(MT)between patients suffered from this kind of arsenism and the normal people was compared and to study the gene expression of MT and its significance.
      Methods   RNA was extracted by the TRIzol Phenol Chloroform method.Quantity of the RNA was determined by UV spectrophotometry and the purity of the extracted RNA was judged by the ratios of A260/A280.The real time-RT-PCR technique was used to mesure the mRNA expression of MT in the coal burntar senism.The actin was used as quality control in the Real time RTPCR.There was statistical difference when the p value was less than 0.05.
      Results   The normal groups mRNA relative expression level of MT was equal to 6.82±7.80.The arsenism groups mRNA relative expression level of MT was equal to 2.02±2.82.The mRNA expression of MT reduced in the patients, and it was approximately equal to 30% of the normal controls.There was statistical difference(P < 0.01)between them.
      Conclusion   The MT has important significance in the metabolism of coal burnt arsenism and plays an improtant role in regulation.The decrease of the mRNA expression of MT in the patients with coal burn arsenism may be related to the aresnism mechanism.
