PTHrP1-34片段对去卵巢大鼠骨质疏松作用

Effect of human Parathyroid Hormone Related Protein(PTHrP1-34)on osteoporosis of ovariectomized rats

      目的   探讨人甲状旁腺激素相关蛋白1-34片段(human parathyroid hormone related protein, PTHrP1-34)对去卵巢骨质疏松大鼠的作用。
      方法   60只4月龄Wistar健康雌性大鼠, 40只行双侧卵巢摘除术, 20只做假手术, 6周后各处死10只证实骨质疏松造模成功。余30只骨质疏松模型鼠随机分为PTHrP治疗组、雌二醇治疗组(E2组)和安慰剂组(Placebo组), 10只假手术组作对照。治疗3个月后, 测定并比较股骨、股椎骨密度(BMD)、血清钙、磷、感性磷酸酶(ALP)、雌激素(E2)、肿瘤坏死因子-α(TNF-α)、骨钙素(BGP)、胰岛素样生长因了(IGF-1)水平。
      结果   治疗后, PTHrP股有和腰椎BMD较Placebo组明显升高; 血清BGP及IGF-1水平也明显高于Placebo组; PTHrP治疗组血清TNF-α无明显变化。
      结论   hPTHrP1-34能通过刺激成骨细胞的活性, 增加骨密度, 对去卵巢大鼠所引起的骨质疏松有效。

     

    Abstract:
      Objective   To investigate the effects of human parathyroid hormone related protein(PTHrP1-34)on the osteoporosis of ovariectomized rats.
      Methods   60 female 4-mont-hold Wistar rats were used for this experiment and 40 of them were ovariectomized and another 20 received sham operation.After 6 weeks of ovariectomy the osteoporosis model was confirmed by use of 10 ovariectomized and sham-operated rats.The osteoporotic rats of 30 were randomly divided into 3 treatment groups, i.e.PTHrP, Estradiol and Placebo.The bone mineral density(BMD), serum Ca, P, alkaline phosphatase(ALP), tumor necrosis facto-rA(TNF-α), bone-carboxy glutamate protein(BGP)and I nsulin-like growth factor-Ⅰ(IGF-1)were measured after 3 months therapy.
      Results   After 12 weeks.treatment the femoral and lumbar BMD in the PTHrP group were increased obviously compared with those of Placebo groups.After treated with PTHrP, serum BGP and IGF-1 significantly increased compared with those of Placebo groups, and serum TNF-αhadn't any change.
      Conclusion   PTHrP1-34 can stimulate osteoblast activity, promote bone mass effect and is effective on protection against osteoporosis.
