苯并(a)芘对小鼠肝细胞凋亡的影响

Effects of BaP on apoptosis of liver cells in mice

      目的   探讨苯并(a)芘(BaP)对小鼠肝细胞Fas/Fas-L凋亡信号通路的影响。
      方法   ICR小鼠灌胃染毒, 染毒剂量分别为0, 5, 10, 20, 40 mg/kg, 采用免疫组织化学法检测肝细胞Fas、Fas-L、caspase-3基因表达情况, 应用吖啶橙/溴化乙啶(AO/EB)荧光双染法检测肝细胞凋亡情况。
      结果   BaP各剂量染毒组肝细胞凋亡发生率及Fas、Fas-L、caspase-3基因表达阳性率与对照组比较差异均有统计学意义(P < 0.05)。
      结论   BaP可诱发小鼠肝细胞发生凋亡, Fas/Fas-L信号通路改变是引起肝细胞凋亡发生的重要途径之一。

     

    Abstract:
      Objective   To study the effects of BaP on apoptosis through the Fas/Fas-Lsignal pathway in mice liver cells.
      Methods   ICR mice were divided into five groups, control group and BaP treated group, which were oral exposure withoil and BaP(5, 10, 20, 40 mg/kg)respectively.Immunohistochemical method was utilized to detect the expressions of Fas, Fas-L, caspase-3 and the apoptosis were measured by AO/EB double stained method.
      Results   The expressions of Fas, Fas-L, caspase-3 and apoptosis rate in BaP treated group were significantly higher than those in control group(P < 0.05).
      Conclusion   BaP could induce apoptosis in mice liver cells and the Fas/Fas-Lsignal pathway was one of the important pathways.
