广东地区人禽流感H5N1血凝素基因特征与进化

Characteristic and evolution on HA gene of human H5N1 avian influenza

  • 摘要: 目的通过对广东地区人禽流感H5N1毒株(HA)基因序列的变异分析,揭示毒株HA基因变异与进化。方法对广东地区人禽流感H5N1毒株(A/GD/01/06)HA基因测序,同时检索全球各地人禽流感H5N1毒株HA基因,采用SPSS 11.0和DNAStar 5.0软件对检索的人禽流感H5N1毒株HA基因核苷酸序列进行聚类、比对和分析;并结合临床资料对变异毒株进行进化速度分析。结果1997~2006年,42毒株HA基因序列聚类分成3类;HA基因89氨基酸位点置换,占15.7%(89/568);2003~2006年H5N1,毒株通过氨基酸第170~172位(NST/S)位点的置换,增加一个糖基化位点。同义变异中,HA基因Ks为1.99×10-5~2.58×10-5,生物进化线性回归方程为Y=4.8×10-9X+1.048×10-5;同时各毒株Ks均大于Ka,进化检验Ks/Ka值显示HA基因进化压力主要来自自然变异。42株H5N1毒株可以分3条进化途径,1997~1998年毒株为1条,2003~2005年东南亚毒株为第2条途径,2005~2006年中国毒株为第3条途径。毒株HK-213-03氨基酸变异显示其变异的进化过度性,而2004年毒株TL-L2004-04氨基酸位点变异值得注意。结论2003~2006年人禽流感H5N1毒株HA基因抗原性与1997年毒株有较大不同,人禽流感H5N1,毒株增加一个糖蛋白位点可能改变毒株致病性;少数毒株氨基酸位点变异较大,值得关注。人禽流感H5N1毒株在自然界变异频繁,但受到鸟禽和人体的免疫压力较小;但随着H5N1,毒株自然进化,H5N1,毒株具有人-人传播能力的概率较大。建议在研制人禽流感H5N1毒株疫苗时,要充分考虑不同毒株HA基因抗原性。

     

    Abstract: ObjectiveTo reveal characteristic and evolution on HA geoe of human H5N1 avian influenza by analyzing variation of the nucleotide sequence of HA gene of human H5Nl avian influenza.MethodsHA gene of Guangdong strain was sequenced and others global HA nucleotide sequences of human H5Nl avian influenza from Internet and their variation were clustered and compared and analyzed with SPSS 11.0 and DNA Star 5.0.ResultsForty-two HA genes of human H5N1 avian influenza in 1997~2006 were clustered into three groups.Eighty~nine amino acids of HA gene were substituted and ac2 counted for 15.7% (89/568).HA gene increased one glycoprotein domain in 170~172 sites(NST/S)in 2003~2006.In the synonymous varimion,Ks in HA gene was between 1.99×10-5~2.58×10-5 and the evolutionary linear regression equation was Y=4.84×10-9 X+1.048×10-5.Meantime,Ks was more than Ka in same strain and Ks/Ka value showed that there was not selective pressure in HA gene evolution.H5N1 strain evolution was divided into three clusters,one for strains in 1997~1998,another for strains in 2003~2005 except for strains from China,third for strain from China in 2005~2006.The variation of strain HK-213-03 showed their transitions between 1997 and 2004,and the variations of the amino acids on HA in TL-L2004-04 was remarkable.ConclusionThere was the differences between HA of strains in 2003~2006 and strains in 1997 and one more glycoprofein domain might increase the viral pathogenicity;it was worth noticing variations of the amino acids on HA in TL-L2004-04.Human H5N1 strains evolve frequently in the natural environment,but were not subject to avian or human immunological pressure.However,the probabilities were great that H5N1 strains possess of humanto human transmittion as the natural evolution.The antigenicity of HA gene was needed to consider in vaccine of different human H5N1 strains.
