天麻素对染铅大鼠脑细胞凋亡影响

Effect of gastrodin on apoptosis of brain cells of rats induced by lead

  • 摘要: 目的探讨天麻素对染铅大鼠脑细胞凋亡及学习记忆功能的影响。方法选择健康雄性Wistar大鼠50只,随机分为5组,分别为实验1,2,3,4组和对照组。除对照组外,各组大鼠均给予0.2%醋酸铅水溶液自由饮用,实验2,3,4组给予天麻素溶液灌胃,浓度分别为100,200,400 ng/kg。用Y型迷宫测试学习记忆,用流式细胞仪测定脑细胞凋亡率。结果(1)在Y型迷宫试验中,实验1组大鼠受电击次数显著高于实验2,3,4组和对照组(P<0.01);大鼠学习受电击次数与天麻剂量呈明显负相关。(2)实验1组脑细胞(包括海马、小脑和大脑皮层)凋亡率显著高于实验2,3,4组和对照组(P<0.01);大鼠脑细胞凋亡率与天麻素剂量呈明显负相关。结论天麻素对染铅大鼠脑细胞凋亡和学习记忆功能损害具有改善作用。

     

    Abstract: ObjectiveTo explore the effect of gastrodin on the apoptosis of brain cells and the impairment of learning and memory induced by lead in rats.Methods50 male Wistar rats were chosen and random divided into 5 groups,respectively;1,2,3,4 and control,10 rats in every group.Except for control all rats were given solution of 0.2% lead acetate,and at the same time group 2,3 and 4 were give gastrodin at the dosage of 100,200 and 400mg/kg respecitively.The ability of learining and memory of each group was measured by Y-maze test,The apoptosis of brain cells was determinged by flow cytometry(FCM).Result(1)The number of electric shocks of group1 was significantly increased compared with 2,3,4and control in Y-maze test(P<0.01);and there was a significant dose-response relationship between electric shocks and gastrodin.(2)the apoptosis of brain cells(P<0.01);and a signficant dose-response relationship between apoptosis and gastrodin was found.ConclusionGastr odin may improve the apoptosis of brain cells and the impairment of learning and memory induced by lead.
