微波辐射后大鼠海马差异表达基因筛选

Screening for differentially expressed genes in rats’hippocampus after microwave radiation

  • 摘要: 目的 研究微波辐射后大鼠海马差异表达基因,以探讨微波辐射致脑损伤的分子机制。方法 采用大鼠寡聚核苷酸芯片筛选30mW/cm2微波辐射后大鼠海马差异表达基因,按照国际通用分类标准对结果进行功能分类;逆转录-聚合酶链反应(RT-PCR)验证部分基因于辐射前后表达差异,并分析相关基因在微波辐射损伤中作用。结果 30mW/cm2微波辐射后6h,大鼠海马中筛选到13个差异表达基因,其中4个表达上调,9个表达下调;辐射后7d差异表达基因为20个,其中4个表达上调,16个表达下调。RT-PCR验证结果与芯片一致。差异表达基因功能涉及应激、转运、代谢、发育分化、细胞凋亡、信号转导、转录、细胞粘附等,并与线粒体损伤、N-甲基-D-天冬氨酸受体(NMDAR)信号通路开放、神经递质释放障碍、膜损伤等密切相关。结论 微波辐射可使大鼠海马多个基因差异表达,为进一步研究微波辐射致脑损伤的机制提供新思路。

     

    Abstract: Objective To search differential gene expression in rats' hippocampus and to explore the molecular mechanism of brain injury caused by microwave.Methods The oligonucleotide gene chip of rat was used to screen for gene expression in rats' hippocampus after microwave radiation at an average power density of 30 mW/cm2.Genes were functional classified by international standard.R T-PCR was used to identify.The interesting genes,and these genes were analyzed in the iniury of microwave.Results There were 13 genes differentially expressed after 6-hour radiation,4 genes up-regulated and 9 genes down-regulated,while 20 genes differentially expressed after 7-day radiation,4 genes up-regulated and 16 genes down-regulated.RT-PCR confirmed the results of gene chips.The genes were involved in metabolism,transport,signal,apoptosis,cell adhesion,transcription regulator,development,differentiation and so on.Conclusion There were some genes differentially expressed in rats' hippocampus induced by microwave,which provided new experimental references for the mechanism of microwave research.
