二巯丙磺钠对溴苯腈致小鼠脏器损伤作用

Effects of Na-DMPS on LD50 and pathological changes of tissues in mouse during toxicity of bromoxynil

  • 摘要: 目的 探讨二巯基丙磺钠(Sodium Dimercaptopropane Sulfonate,Na-DMPS)对溴苯腈急性中毒小鼠半数致死量(LD50)及主要脏器病理结构的影响。方法 按Bliss法,计算溴苯腈急性中毒组和二巯基丙磺钠保护组小鼠半数致死量,并观察2组小鼠脏器病理变化。结果 二巯基丙磺钠保护组小鼠半数致死量显著高于溴苯腈急性中毒组(P<0.01)。光镜可见溴苯腈急性中毒小鼠肝充血明显;脑组织肿胀,脑实质中可见胶质小结。二巯基丙磺钠保护组小鼠组织无明显损伤。电镜可见溴苯腈急性中毒小鼠肝细胞内质网扩张,少量线粒体气球样变性,部分核固缩;肺组织内皮细胞水肿;大脑皮质有髓鞘样小体形成,神经微丝微管消失。二巯基丙磺钠保护组小鼠肝细胞仅见内质网扩张;脑皮质仅见部分神经突起水肿。结论 溴苯腈急性中毒组小鼠出现多脏器功能哀竭,二巯基丙磺钠能显著提高溴苯腈急性中毒小鼠半数致死量,并可减轻主要脏器病理损伤。

     

    Abstract: Objective To demonstr ate the effects of Na-DMPS on the LD50 and pathological changes of the main tissues in mouse during acute toxicity of br omoxynil.Methods Using Bliss method to calculate the LD50 of bromoxynil of the group of mice in acute bromoxynil poisoning and the group of mice in acute bromoxynil poisoning after Na-DMPS protection,and to observe the pathological changes of the main tissues of the two groups of mouse under optical microscope and electron microscope.Results The LD50 of the group of mice in acute bromoxynil poisoning after Na-DMPS protection increased much more obviously than the LD50 of the group of mice in aute bromoxynil poisoning(P<0.01).Under optical microscope:hyperemia in every lobe was obvious;there was swelling in brain,some colloid nodes could be seen in brain substance.But tissue damage in the group of mice in acute bromo xynil poisoning after Na-DMPS protection was not obvious.Under electron microscope,the group of mice in acute bromoxynil poisoning:dilation of endoplasmic retculum,balloning degeneration of mitochondrion and pyknosis were observed;there was swelling in endotheliocytes of lung;neural sheath was observed,nerve microfilament and microbule disappeared.A bout the guoup of mice in acute bromoxynil poisoning after Na-DMPS protection:dilation of endoplasmic reticulum in liver cell was observed;Edema could be seen in part of cerebral cortex.Conclusion The group of mice in acute bromoxynil poisoning shows multiple organ disfunction syndrome.The protection of Na-DMPS can increase the LD50 of bromox ynil obviously and lessen pathological changes of the main tissues.
