Effect of Schistosoma japonicum sj23 gene transfer on function of dendritic cell in vitro

  • 摘要: 目的 探讨日本血吸虫23kDa膜蛋白(Sj23)基因转染对树突状细胞(DC)功能的影响。方法 利用脂质体介导的基因转染技术将Si23基因转染DC,流式细胞术(FCM)检测Sj23基因转染的DC摄取抗原的能力,混合淋巴细胞反应(MLR)检测Sj23基因转染的DC对同种异体T淋巴细胞的刺激作用。结果 Sj23基因转染后Sj23蛋白在DC中高表达,与pcDNA3转染的DC和未转染的DC相比,Sj23基因转染的DC摄取抗原的能力降低,能明显刺激混合淋巴细胞反应。结论 Sj23基因转染DC后能促进DC的活化,增强DC的生物学活性。

     

    Abstract: Objective To explore the effect of 23kDa membrane protein of Schistosoma japonicum(Sj23)gene transfer on the function of dendritic cell(DC)in vitro.Methods Sj23 gene was introduced into DC by liposome-mediated gene transfer met hod.DCs were transfected with recombinant plasmid pcDNA3-Sj23.The ability of DC to take an antigen was determined by flow cytometry(FCM).The mixed lymphocy tereaction(MLR)was used to detect the effect of Sj23 gene-transferred DC on stimulating allogenetic Tlymphocyte.Results Sj23 was expressed at high level in the Sj23 gene-transferred DC.Compared with pcDNA3 transfer red DC and untreated DC,the ability of antigen intake of Sj23 gene-transfer red DC decreased and Sj23 gene-transferred DC had strong potential to stimulate proliferation of allogenetic Tlymphocy te.Conclusion Sj23 gene-transfer can promote DC activation and enhance the biologic activity of DC.
