苦参碱对体外白念珠菌生物膜抑制作用

In vitro activity of matrine against Candida albicans biofilms

  • 摘要: 目的 研究苦参碱对体外白念珠菌生物膜的影响.方法 采用甲基四氮盐(XTT)减低法评价苦参碱对白念珠菌的生物膜及黏附牲的影响.镜下观察苦参碱对白念珠菌生物膜的形态学影响;细胞毒试验检测苦参碱毒副作用.结果 苦参碱抑制白念珠菌生物膜50%及80%的药物浓度(SMiC50、SMIC80)分别为500,1000μg/ml.1000μg/ml苦参碱对培养0,1,2,4h的白念珠菌黏附,100μg/ml苦参碱对培养0,1h的白念珠菌黏附;10μg/ml苦参碱对培养0h的白念珠菌黏附均有抑制作用;1000μg/ml苦参碱完全抑制白念珠菌生物膜的形成,且只引起14.05%红细胞溶血.结论 苦参碱对体外白念珠菌生物膜有较强的抑制作用.

     

    Abstract: Objective To investigate in vitro effects of matrine on C.albicans biofilms.Methods XTT reduction assay was performed to determine the effect of matrine on C.albicans biofilms and its adherence.T he effect o f matrine on morphog enesis of C.albicans biofilms was obser ved by inverted microscopy,and cytotox ic assay was used to measure the adverse effects of matrine.Results SMIC50,SMI C80 of matrine against C.albicans biofilms was 500,1000 Lg/ml,respectively.M atrine at the concentration of 1000 Lg/ml could inhibit the adherence of C.albicans at 0,1,2,4 h.M atrine at the concentration of 100 Lg/ml could inhibit the adherence at 0,1 h,and 10 Lg/ml,at 0 h,respectively.1000 Lg/ml of matr ine could inhibit biofilm formation,and cause 14.05% of hemolysis.Conclusion M arine shows potential activ ity ag ainst C.albicans biofilm in the study.
