内毒素耐受对D-GalN/LPS致大鼠肝损伤保护

Effect of endotoxin tolerance on D-GalN/LPS induced fine structure change of liver and cytokine in rats

  • 摘要: 目的 研究内毒素耐受(endotoxin tolerance,ET)对大鼠接受D-胺基半乳糖(D-Gal N)/脂多糖(LPS)刺激后肝组织超微结构改变及血清细胞因子变化的影响.方法 雄性SD大鼠先以0.1mg/kg LPS腹腔注射,每日1次,连续5次,于第5次注射24h后再腹腔注射D-Gal N800mg/kg和LPS8μg/只,注射D-Gal N/LPS24h后留取大鼠血及肝脏标本.苏木素-伊红染色及透射电镜下观察肝组织病理及超微结构变化;采用鲎试剂基质显色法测定血清内毒素水平,ELISA法检测肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)水平.结果 ET组大鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBiL)水平均明显低于急性肝衰竭(ALF)组(P<0.01),且ET组大鼠肝组织病理改变及超微结构变化明显轻于ALF组;同时,ET组大鼠血清内毒素、TNF-α、IL-6水平亦明显低于ALF组(P<0.01).结论 内毒素耐受时,大剂量的D-Gal N和LPS腹腔注射可减轻肝组织损害,内毒素、TNF-α、IL-6释放减少,提示内毒素耐受可能有助于防治急性肝功能衰竭.

     

    Abstract: Objective To study the fine structure changes of liver and the level of cytokine in endotoxin tolerant rats received intraperitoneal injections of GalN/LPS.Methods Endotoxin tolerance(ET)model of SD rats was induced by five daily intraperitoneal injections of 011mg/kg LPS.Normal control rats received intraperitoneal injections of the same volume saline.Then the rats were injected with D-GalN 800 mg/Kg and LPS 8μg/rat.On the sixth day,liver tissue and blood were collected 24 hours after the injection of D-GalN/LPS.Liver histopathology and fine structure of each group were observed by HE staining and electron microscope.The level of TNF-αand IL-6 were estimated by ELISA and the concentrations of endotoxin were determined by tachypleus amebocyte lysate.Results GalN/LPS induced acute liver injury was attenuated significantly by LPS pretreatment.The plasma ALT,AST and TBiL activity were decreased remarkably by LPS pretreatment.The concentrations of endotoxin and the TNF-αand IL-6 level were evidently lower in ET group than those in the ALF group.Conclusion LPS pretreatment could induce endotoxin tolerance of rat,which made decreased releases of TNF α,IL-6 and endotoxin.The alleviation of GalN/LPSinduced acute liver injury in ET rats suggested that ET could be used to reduce severe liver injury.
