高血压大鼠肾脏中HMGCS2表达及抗氧化作用

Different renal HMGCS2 expressions between spontaneously hypertensive rat (SHR) strains and its antioxidative role

  • 摘要: 目的 探讨自发性高血压大鼠(SHR)和卒中易感型自发性高血压大鼠(SHRSP)肾脏组织中3-羟基-3-甲基戊二酰辅酶A合成酶2(HMGCS2)的表达差异及其在抗氧化中的作用。方法 随机选取2月龄雄性SHR和SHRSP大鼠各50只,用免疫印迹法检测肾组织中HMGCS2和过氧化氢酶表达差异以及细胞信号磷酸化水平;同时测量组织中丙二醛(MDA)含量。结果 SHRSP组HMGCS2表达量(6 398±658.128)明显低于SHR组(16516.67±1321.855)(P<0.05)。SHRSP组MDA含量(0.738±0.175)U/mg prot高于SHR组(0.330±0.032)U/mg prot(P<0.05);SHRSP组蛋白激酶B(Akt)的磷酸化水平(1.909±0.124)明显高于SHR组(0.229±0.071)(P<0.05)。结论 HMGCS2在SHRSP和SHR肾脏的表达差异有统计学意义,并可以在抗氧化中发挥作用;其表达可能由磷酸化蛋白激酶B(p-Akt)信号调节。

     

    Abstract: Objective To compare the differet expressions of 3-hydroxy-3-methyl glutary l-CoA,synthase 2 (HMGCS2)in kidney bewteen stroke prone spon taneously hypertensive rat(SHRSP)and spontaneo usly hypertensive rat (SHR)strains and explore its antiox idative role.Methods Five male SHRSP and five male SHR at the age of 2 months were randomly chosen.The expressions of HMGCS2,cata lase and phosphory lating signal of prote in kinase B(Akt)pathway were evaluated by mimunoblotting analysis.The level of malond ialdehyde(MDA)was also measured.Results Lower renal HMGCS2 expre ssion(6398±658.128 v s16516.67±1321.855,P<0.05),higher MDA level(0.738±0.175 U/rag prot vs 0.330±0.032 U/mg prot,P<0.05)and higher Akt phosphorylation signal(1.909±0.124 vs 0.229±0.071,P<0.05)in SHRSP than those of in SHR were observed.Conclusion The different expression of renal HMGCS2 between SHRSP and SHR may be partially responsible for the an tioxida tive property.
