广州市儿童秋冬季诺如病毒感染检测分析

Detection of Norovirus among children in Guangzhou in fall-winter seasons of 2003-2005

  • 摘要: 目的 研究广东省广州市5岁以下儿童秋冬季急性病毒性腹泻诺如病毒的感染情况、基因型别和流行优势毒株。方法 采用逆转录-聚合酶链反应(RT-PCR)方法检测诺如病毒(NVs),部分阳性标本的PCR产物经纯化、测序,结合参考毒株相应的核苷酸序列进行进化分析。结果 NVs的检出率为23.88%(112/469)。35株测序病毒之间的核苷酸序列同源性为70.4%~100%,均属于GII遗传组。广州市2003年秋冬季测序的11株NVs 5株为GII-4;5株为GII-3;另1株不能确定其型别;2004年测序的11株10株为GII-4;1株为GII-3;2005年所测序的13株均为GII-4。结论 广州市婴幼儿秋冬季病毒性腹泻存在不同基因型NVs感染,以NV GII组毒株为主,其中GII-4型为流行优势株。

     

    Abstract: Objective To investigate Noroviruses (NVs) infection among children under 5 years of age with nonbacterial diarrhea.Methods NVs were detected by amplification of cDNA from the viral RNA by RT-PCR using priner pair JV13I/JV12Y.Amplicons obtained from NVs-positive stools were purified and sequenced Sequences were compared with prototye sequences retrieved from NCBI database.Results 957 fecal specie ens were collected.For 469 rotavirus-negative specieens,NVs positive rate was 23.88%(112/469).All 35 sequenced strains belonged to NLV CII in which 28 strains were CII-4,6 strains were CII-3 and 1 strain could not be d identified.Conclusion NVs with different genotypes circulated among children in Guangzhou,and CII-4 were the prevalent strain.
