六月青乙醇提取物对鸭乙型肝炎病毒抑制作用

Inhibitory effect of ethanol extract of Liuyueqing on heoatitis B virus in duck

  • 摘要: 目的观察六月青乙醇提取物(EELYQ)对鸭乙型肝炎病毒乙型肝炎表面抗原(DHBsAg)和乙型肝炎E抗原(DHBeAg)的抑制作用。方法采用1日龄雏鸭接种广西麻鸭乙型肝炎病毒(DHBV)强阳性血清,13 d后用PCR法筛选出DHBV强阳性鸭。随机分成5组:EELYQ高剂量组、中剂量组、低剂量组、模型组和阳性对照组,每组10只,各组雏鸭均灌胃给药14 d。于用药前(T0)、用药7 d(T7)、14 d(T14)及停药后3 d(P3)分别采血,采用ELISA法测定上清液DHBsAg和DHBeAg的滴度。结果EELYQ中、高剂量组给药后血清DHBsAg和DHBeAg的滴度显著降低(P<0.01),停药3 d后,EELYO高剂量组血清DHBsAg和DHBeAg滴度无反跳现象。EELYQ高剂量组给药14 d(T14)和停药3 d(P3)后,DHBsAg抑制率分别达到51.85%和59.04%。EELYQ高剂量组给药14 d(T14)和停药3 d(P3)后,DH-BeAg抑制率分别达到34.27%和39.96%。EELYQ抑制DHBsAg和DHBeAg作用有明显的量效和时效反应关系。结论EELYQ可有效抑制DHBsAg和DHBeAg的表达。

     

    Abstract: ObjectiveTo study the inhibitory effect of ethanol extract of Liuyueqing(EELYQ)on duck hepatitis B surface-antigen(DHBsAg)and hepatitis B e-antigen(DHBeAg).MethodsOne day old Guangxi brown spotted ducks in fected with DHBV were used as the hepatitis B virus infection animal model.DHBsAg and hepatitis B positive ducks were detected by PCRat 13 day after stopping the infection of DHBV,and were randomly divided into five groups:the high,middle and low dose EELYQ groups,model group,positive control group.Each group had 10 ducks.EELYQ was given.i g.for 14 days,and the contents of DHBsAg and DHBeAg in serum were measured by ELISA.ResultsThe serum DHBsAg and DHBeAg contents were decreased significantly by the treatment of EELYQ.The high and middle dose of EELYQ could significantly suppress DHBsAg and DHBeAg expressions in vivo(P<0.01).DHBsAg and DHBeAg contents in serum of high dose group did not return significantly 3 days after stopping treatment.The DHBsAg inhibition rate was 51.85% and 59.04%,respectively at 14 days treatment and 3 days after stopping the treatment in high EELYQ dose group and for DHBeAg,the inhibition rate was 34.27% and 39.96%,respectively.The inhibitory effect of EELYQ was dose and time dependent.ConclusionEELYQ can significantly inhibit the expressions of DHBsAg and DHBeAg.
