猪链球菌2型多重PCR鉴定方法建立

A multiplex PCR assay application in identification of Streptococcus suis type 2

摘要: 目的建立猪链球菌2型多重PCR鉴定方法。方法根据猪链球菌种特异基因16S rRNA序列和猪链球菌2型特异性荚膜多糖编码基因cps2 J基因序列,设计和合成2对特异引物,通过条件和体系优化,建立多重PCR检测方法。结果猪链球菌2型参考株及18株分离株均扩增到清晰的目的条带,而6株非猪链球菌2型参考菌株不能扩增出目的条带。结论多重PCR可以用于猪链球菌2型检测,特异性和敏感性好,为该病的快速诊断和分子流行病学调查提供了新的技术方法。

Abstract: ObjectiveTo develop a rapid identification method for Streptococcus suis type 2 with multiplex PCR assay.MethodsAccording to specific sequence of Streptococcus suis type 2, two pairs of multiplex PCR priners were designed for Streptococcus suis amplification and identification.Then the amplification conditions were optinized.ResultsThe ain gene fragment for 17 isolates and 1 reference of Streptococcus suis type 2 were amplified with multiplex PCR assay, but it could not be amplified for 6 other reference bacteria.ConclusionThe multiplex PCR assay could be used to identify and provide a new methodology for Streptococcus suis type 2 identification.
