菜市场污水中细菌多样性检测分析

Technique for bacterial detection in food market sewage

  • 摘要: 目的 未经处理的菜市场污水中携带大量细菌(特别是病原细菌),查清菜市场污水中细菌多样性分布和种群结构特征,为菜市场污水治理措施提供基础数据。方法 通过提取水中微生物总DNA,利用PCR扩增细菌16S rRNA基因片段,构建污水中细菌16S rRNA基因文库。结果 菜市场宰杀清洗家禽和水产品产生的污水中细菌多达16个属30多种,细胞16S rRNA基因主要来自变形细菌(proteobacteria)的ε-,γ-,δ-,β-亚族、厚壁菌门(Firmiutes)和拟杆菌门(Bacteroidetes)等类群,近似弓形菌属(Arcobacter)和梭菌属(Clostridium)的细菌比例最高,分别为27.6%和20.7%。结论 构建细菌16SrRNA基因文库的分子生物学方法可快速检测菜市场污水中细菌,且细胞种群呈多样性。

     

    Abstract: Objective To detect pathogenic bacteria in food market sewage.Methods A 16S ribosom al DNA clone library from food market sewage was established by PCR using suitable primers.Results The sequence analysis indicated that the bacteria were mainly iden tified asε-,γ-,δ-,β-proteoba cteria,Firmicutes,Bacte roidetes group.The prop ortion of Arcobacter and Clostridium were 27.16% and 20.7%,respectively.Conclusion The results proved that there were abundant bacteria in food market sewage.The molecular biologic methodes tablished can rapidly detect the bacte ria in the sewage.
