枸杞多糖对染铅小鼠学习记忆及海马区PKC影响

Influenec of Lycium barbarum polyccharides on learning and menory function and protein kinase C in hippocampus in lead-exposed mice

  • 摘要: 目的 探讨枸杞多糖对染铅小鼠学习记忆功能和海马区蛋白激酶C(Protein kinase C,PKC)的影响。方法 将实验小鼠随机分为5组,即空白对照组、染铅模型组、枸杞多糖高、中、低3个剂量组,枸杞多糖各剂量组于染铅后第2d灌胃给药,每天1次,于第2,4周测小鼠到达迷宫避难台所需要的时间,第4周测血铅浓度和海马区PKC含量。结果 枸杞多糖各剂量组与染铅模型组比较,血铅浓度枸杞多糖各剂量组(0.65±0.28)~(0.11±0.13)mg/L,模型组(1.68±0.19)mg/L明显降低,到达避难台的时间也明显减少;而海马区PKC含量枸杞多糖各剂量组(73.23±6.52)~(96.33±8.03)mmol/L,模型组(62.26±7.21)mmol/L明显升高,差异有统计学意义(P<0.05或P<0.01)。结论 枸杞多糖可促进小鼠体内铅的排出,改善染铅小鼠的学习记忆功能,抑制海马区PKC含量降低。

     

    Abstract: Objective To explore the influence of Lycium barbarum polyccharides(LBP)on learning and memory function and the content of protein kinase C(PKC)in hippo campus of lead-exposed mice.Methods The mice were divided randomly into five groups:control group,lead-exposed model group,high-,mid-and low-dosage LBP groups.The LBP groups were fed with different do sage of LBP since the second day of the lead exposure.Blood-lead concentration,time of reaching water maze platformat 2nd and 4 th week,and the concentration of PKC in hippo campuswrere detected.Results Compared with lead-expo sure model group,the blood-lead concentrations of the mice in LBP groups was lower (0.65±0.28-0.11±0.13 mg/L vs 0.68±0.19mg/L)and the tmie of reaching at the platform was less.The concentration of PKC in LBP groups(73.23±6.52-96.33±8.03 mmol/L)was higher than that of in lead-explsed model guoup (62.26±7.21 mmol/L).All the diffenrences were statistically significant(P<0.05).Conclusion LBP promotes the discharge of lead and improves learning and memory function,and inhibits PKC decrease in hippocam pusin lead-exposed mice.
