氯化三丁基锡致巨噬细胞凋亡作用

Apoptosis of RAW 264.7 cells induced by tributyltin chloride

  • 摘要: 目的 探讨氯化三丁基锡(tributyltin chloride,TBTCl)致巨噬细胞株RAW264.7细胞的凋亡作用。方法 当RAW264.7细胞处于代数生长期时,分别加入不同浓度(0.0,0.025,0.05,0.10,0.20,0.40mg/L)的TBTCl处理48h,分别用四甲基偶氮噻唑蓝(MTT)法检测细胞活性,观察TBTCl对RAW264.7细胞增殖影响;吖啶橙/溴化乙锭(AO/EB)染色法观察凋亡细胞的形态,流式细胞仪检测TBTCl对RAW264.7细胞的凋亡率。结果 TBTCl能抑制RAW264.7细胞的增殖,其半数抑制浓度(IC50)=0.179mg/L;TBTCl也能诱导细胞的凋亡,当TBTC1达到0.40mg/L时,细胞凋亡率达到19.5%。结论 TBTC1可诱导细胞凋亡。

     

    Abstract: Objective To explore the apoptosis effect of tributyltin chloride(TBTC1)on RAW 264.7 cells.Methods RAW 264.7 cells were incubated with various concentrations of TBTCL for 24 hours.The cell growth inhibition wasm easured by MTT,and the cell apoptosis was observed by flow cytometry analysis and fluo rescence micro scope.Results TBTC l could result in the descent of cell activity(IC50=0.179 mg/L)and could induce the apoptosis in RAW 264.7 cells.The rate of apoptosis of RAW 264.7 cells reached to 19.5% at TBTC l concentration of 0.4 mg/L.Conclusion TBTC l could induce apoptosis of RAW 264.7 cell.
