广西巴马瑶族STR基因座遗传多态性分析

Genetic polymorphisms of STR loci in Bama Yao ethnic group of Guangxi

  • 摘要: 目的分析广西壮族自治区巴马瑶族人群15个短串联重复序列基因座(STR)遗传多态性。方法采用聚合酶链反应-短串联重复序列(PCR-STR)技术及遗传分析仪,对巴马瑶族114名个体血样DNA样品进行各基因座等位基因频率及杂合度(H)、个体识别力(DP)、非父排除率(EP)、多态信息含量(PIC)等群体遗传学指标分析。结果15个基因座共检出等位基因120种,频率为0.004 4~0.530 7。除TPOX基因座外,其余14个位点的H为0.6228~0.868 4,PIC为0.592 9~0.844 1,DP为0.817 6~0.959 1,EP为0.319 1~0.731 5;累积个体识别力达0.999 999 999,累积非父排除率达0.999 953 702。巴马瑶族分别与金秀瑶族、临桂瑶族、清远瑶族比较,15个STR基因座大部分等位基因频率分布差异均有统计学意义(P<0.05)。结论广西巴马瑶族的14个基因座属于高度多态性遗传标记,可应用于民族群体遗传学研究。

     

    Abstract: ObjectiveTo study the genetic polymorphismof 15 short tandem repeats(STRs)loci in Bama Yao ethnic group in Guangxi.MethodsDNA samples from 114 Bama Yao unrelated individuals in Guangxi were analyzed with multi-plex PCR amplification and Genetic Analyzer.Gene frequency,heterozygosity(H),discrimination power(DP),probability of paternity exclusion(EP)and polymorphism information content(PIC)were calculated.ResultsTotally 120 alleles in 15 STR sloci were observed with gene frequency of 0.0044-0.5307.The H was at 0.6228-0.8684,PIC was at 0.5929-0.8441,DP was at 0.8176-0.9591,and EP was at 0.3191-0.7315 in observed STR sloci except TPOX.The cumulative paternity of exclusion power for the seSTR markers in Bama Yao population was 0.999 953 702 and the cumulative discrimination power was 0.999 999 999(except TPOX).Compared with Jinxiu Yao,Lingui Yao,and Qingyuan Yao ethnic groups,there were significant differences between Bama Yao ethnic group and the three groups in allele frequencies at most of 15 STR sloci.ConclusionThe 14 STR sloci of Bama Yao ethnic group in Guangxi show high genetic polymorphism.They can be used as genetic markers of Bama Yao ethnic group in population genetics study.
