Abstract:
ObjectiveTo investigate protective effects of A-lipoic acid(α-LA)on glycome tabolism enzymes in rathepatoctes damaged by H
2O
2.
MethodsAfter exposed to α-LA of 50,100,and 200 LM,respectively,for 24 hr,rathepatocytes were damaged by H
2O
2 for 1 hr.Then the activity of cells,the activity of superoxide diamutase(SOD)and glutathione peroxidase(GSH-Px),maleic dialdehyde(MDA)levels,and the contents of glucokina se(GK)and glycogen syntha sekinase-3(GSK-3)were detected.
ResultsThe cell activity,the activity of GSH-Px(94.211±3.958 U/mg·pro)and SOD(9.636±0.480 U/mg·prot)in H
2O
2 exposure group were lower than those of in the control group(
P<0.05).The levels of MDA in H
2O
2 exposure groups were higher than that of in control guoup.And the contents of MDA in every α-LA group(14.142±0.377,12.064±1.739,and 13.966±0.520 nmol/mg·prot)were lower than that of in the H
2O
2 group (20.435±1.739 nmol/mg·prot,
P<0.05).The content of GK(0.677±0.111 mg/L)in the H
2O
2 group was lower than that of in the control group(1.476±0.280 mg/L).The content of GK in low-dose α-LA group(1.020±0.115 mg/L) was higher than that of in the H
2O
2 group(
P<0.05).
Conclusionα-LA could prevent the oxidative damage of ratheptocytes induced by hydrogen peroxid and could increse the content of GK in rats.
