δ-生育三烯酚对人结肠癌SW620细胞抑制作用

Anti-proliferative effect of δ-tocotrienol on colorectal cancer cells

  • 摘要: 目的 探讨δ-生育三烯酚对人结肠癌SW620细胞增殖抑制作用.方法 采用细胞生长曲线和real-time RT-PCR方法,观察各剂量下δ-生育三烯酚对细胞增殖抑制及对丝/苏氨酸磷酸激酶(GSK-3β)、结肠癌腺瘤性息肉蛋白(APC)及轴蛋白(Axin)在mRNA水平上表达的影响.结果 δ-生育三烯酚对人结肠癌SW620细胞有明显抑制作用,24 h细胞生存率为14.4%~88.8%;GSK-3β及APC mRNA表达水平无明显变化,Axin mRNA表达水平为2.24~2.58,明显高于对照组(P<0.05).结论 δ-生育三烯酚能够明显抑制SW620细胞的生长,其机制可能与上调Axin mRNA水平有关.

     

    Abstract: Objective To investigate inhibitory effect of δ-tocotrienol on prolife ration of colorectal cancer(CRC) SW 620 cells.Methods SW620 cells we retreated with different concentration of δ-tocotrienol(0,20,30,40,60 mol/L).Methyl thiazolylte trazolium(MTT) method was adopted for the detection of cell growth and real time reverse transcription polymerase cha in reaction(RT PCR) for the detection of corre lated mRNA expression.The contents of GSK-3β,APC,and Ax in mRNA were detected.Results δ-tocotrienol inhibited proliferation of CRC-SW 620 cells.After treated with δ-tocotrienol for 24 hr,the survival rate of the cells was decreased by 14.4% to 88.8%.The RT PCR study showed that δ-tocotrienol promoted the expression of Ax in mRNA(F=6.598,P < 0.05),but not the GSK-3β mRNA and APC mRNA(P >0.05).Conclusion δ-tocotrienol can inhibit SW 620 cells proliferation.Its inhibitory mechanism correlates possibly with the adjusting of the associated expression of Ax in mRN A.
