日本血吸虫抗体动态变化SAS软件分析

Analysis on dynamic change of specific IgG antibody induced by schistosomiasis japonica soluble egg antigen with PROC MIXED procedure in SAS

  • 摘要: 目的 应用SAS软件对日本血吸虫感染小鼠血清中可溶性虫卵抗原(SEA)特异性抗体进行动态趋势分析.方法 将2种品系小鼠(BALB/c和C 57 BL/6)分别分成3组,感染、治疗和重复感染组,通过内眦静脉定期采集小鼠血样,采用酶联免疫吸附法(ELISA)测定血清中可溶性虫卵抗原IgG抗体水平;应用SAS软件分析检测结果.结果 自感染日至感染后20 d,日本血吸虫可溶性虫卵特异性抗体IgG无明显变化(P>0.05);感染后30~60 d IgG明显上升,差异有统计学意义(P<0.01);感染后70~120 d IgG亦无明显变化(P>0.05).结论 SAS提供的混合线性模型及相应的PROCMIXED过程可以应用于医学数据的相关性分析.

     

    Abstract: Objective To analyze the serumlevel of soluble eggantigen(SEA)induced specific antibody IgG in or derto understand the dynamics of the IgG with PRO C MIX ED procedure in SAS.Methods Thirty female mice aged 6-8 weeks were divided into three groups:infection group(group A),the rapy group(group B),and reinfection grouop(group C).Ten cercarie/permice were used to infect the mice of group A and group B.Group B was treated with praziquantel at the dosage of 400 mg/kg 50 days after the infection.Group C was infected for the second time with 10 cercarie/per mice 70 days after the first infection.We used fiber micro tube to collect blood samples from angularvein of mice and preserve them at-20℃ frozen condition.A fterwards,we used enzyme linked mimuno sorbent a ssay(ELISA)to detect seruman tibody levels.Then we used PROC MIX ED procedure in SAS to analyze the fixed effect and ram dom effect of SEA induced spec ific an tibo dy IgG leve.l R esu lts There was no significant difference in IgG antibody at the time between 0 and 20 days a fter the infection.There was also no significant difference in IgG antibody at the time between 70 and 120 days a fter the infection.There were significant differences(P<0.0001)in IgG antibody at the time between 30 and 60 days after the infection.Conclusion The results present a dynamic change of specific antibody IgG induced by schisto somia sisjaponica SEA and coincides well with the biological observatio n.PRO CMIXED procedure could be adoted in the analysis of medical experiment data.
