TIP30在脑星形细胞瘤中表达及意义

Expression of TIP30 protein in brain astrocytoma

  • 摘要: 目的 探讨Tat(transactivator of transcription)结合蛋白30(Tat-interactive protein 30,TIP30)在脑星形细胞瘤发生发展中的作用.方法 采用免疫组化链霉菌抗生物素蛋白过氧化酶法(SP)和末端脱氧核苷酸转移酶介导的缺口末端标记法(TUNEL)分别检测69例脑星形细胞瘤和8例正常脑组织中TIP30蛋白表达、增殖细胞核抗原标记指数(PCNA LI)、微血管密度(MVD)和瘤细胞的凋亡指数(AI).结果 69例脑星形细胞瘤组织TIP30的阳性表达率为49.3%,明显低于正常脑组织100%(P<0.01);TIP30的阳性表达率随星形细胞瘤的病理分级级别的升高而降低(χ2=16.31,P<0.01);TIP30蛋白阳性表达的星形细胞瘤组织中的MVD值(30.08±5.63)和PCNA LI值(22.29±7.15)明显低于TIP30蛋白阴性组的MVD值(47.83±8.27)和PCNA LI值(48.11±7.16)(P均<0.01);TIP30蛋白阳性表达的星形细胞瘤组织中的AI值(6.66±1.03)明显高于TIP30蛋白阴性组的AI值(4.02±1.09)(P<0.01).结论 星形细胞瘤中TIP30蛋白表达下降,可能是通过促进新生血管形成、瘤细胞增生和抑制凋亡作用参与了脑星形细胞瘤的发生发展过程.

     

    Abstract: Objective To explo re putative roles of transactivator of transcricption interactive prote in 30(TIP30)in the initiation and progression of a strocytoma.Methods The expression of TIP30,proliferating cell nuclear an tigen labeling index(PCNA L I),microv ascular density(MVD),apoptosis index(AI)in 69 cases of astrocy tomas and 8 cases of normal braintissue were comparatively studied with immunohistochemical staining method and termindal deoxynuc leotidyl transfernase mediated nick end labeling(TUNEL).Results The expression of TIP30 in astrocy toma was lower than that of normal brain tissue and was negatively related to the histo logical grade of tumor(χ2=16 31,P<0.01).MVD and PCN ALI with positive expression of TIP30 in astrocy toma were significantly lower than that with negative expression(P<0.01,P<0.01).AI with positive expression of TIP30 in astrocy toma was significantly higher than that with negative expression(P<0.01).Conclusion The decrease in expression of TIP 30 may play an important role in the in itiation and prog ression of astrocy tom a by promotingang iogenesis,tumor cell hype rplasia and decreasing apoptosis.
