姜黄素对脂肪细胞脂联素及抵抗素表达影响

Effect of curcumin on expression of adiponectin and resistin mRNA in 3T3-L1 adipocytes

  • 摘要: 目的 探讨姜黄素对3T3-L1脂肪细胞脂联素和抵抗素mRNA表达的影响。方法 在诱导3T3-L1前脂肪细胞分化过程中加入姜黄素,于第6 d收集细胞,或用姜黄素处理分化成熟3T3-L1脂肪细胞,24 h后收集细胞,采用逆转录聚合酶链反应(RT-PCR)方法检测脂肪细胞中脂联素和抵抗素mRNA水平。结果 在3T3-L1前脂肪细胞分化期间,姜黄素2.5,5,10,15和20μmol/L分别使脂联素mRNA表达增强48.7%,25.6%,89.7%,128.2%和207.7%,抵抗素mRNA表达下降了42.3%,35.4%,31.5%,58.3%,33.9%;20μmol/L姜黄素使成熟脂肪细胞脂联素mRNA表达增强203.3%;5,10,20μmol/L的姜黄素分别使抵抗素mRNA的表达降低15.2%,49.0%和55.6%。结论 姜黄素可增强脂肪细胞脂联素mRNA表达,降低抵抗素mRNA表达。

     

    Abstract: Objective To explore the effect of curcumin on the expressions of adiponectin and resistin mRNA in 3T3 L1 adipocytes,and to analyze possible mechanism of curcumin in the prevention of diabetes at cellcular level.Methods Curcumin at concentrations of 2.5,5,10,15,and 20 μmol/L was added in 3T3-L1 preadipocytes during induced-differentia tion for six days and 5,10,and 20μmol/L in mature 3T3-L1 adipocytes for 24 hr,respectively.Then the expressions of adi ponectin and resistin mRNA in 3T3-L1 preadipocytes during induced-differentiation and mature 3T3-L1 adipocytes were de tected with RT-PCR.Results The expression of adiponectin mRNA increased and resistin mRNA decreased in 3T3-L1 preadipocytes treated with curcum in compared to 3T3-L1 preadipocyte swithout curcum in during induced-differentiation.The expression of adiponectin mRNA increased and resistin mRNA decreased in mature 3T3-L1 adipocytes treated with curcum in compared to that of in mature 3T3-L1 adipocytes without curcumin.Conclusion The increase of adiponectin expression mRNA and the decrease of resistin mRN A in 3T3-L1 adipocytes may be one of mechanisms of curcumin in the prevention of diabetes.
