Association of genetic polymorphism of IL-12p40+1188A/C with HCMV infection

  • 摘要: 目的 探讨人巨细胞病毒(HCMV)感染儿童的白细胞介素12(IL-12)亚基p40 3’非翻译区+1188(A/C)位点基因多态性与HCMV感染的关系.方法 应用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)和DNA测序的方法,检测125例HCMV活动性感染儿童、108例HCMV潜伏性感染儿童和110例健康儿童IL-12p40 3’非翻译区+1188(A/C)位点的基因型,采用荧光定量聚合酶链反应(FQ-PCR)测定HCMV活动性感染儿童尿液HCMV DNA载量.结果 HCMV活动性感染组、潜伏性感染组和对照组之间AA、AC、CC基因型分布差异有统计学意义(χ2=10.542,P=0.032);HCMV活动性感染组A、C等位基因分布频率分别为52.8%、47.2%,潜伏性感染组分别为63.4%、36.6%,对照组分别为67.7%、32.3%,3组的差异有统计学意义(χ2=11.777,P=0.003);HCMV活动性感染组与非活动性感染组(潜伏性感染组+对照组)的等位基因分布频率差异有统计学意义(χ2=10.930,P=0.001;ORA=0.805,95%CI:0.703~0.922;ORC=1.372,95%CI:1.141~1.650);不同基因型HCMV活动性感染儿童的尿HCMVDNA载量差异有统计学意义(F=9.325,P=0.006),其中CC基因型患儿尿HCMV DNA载量高于AA基因型(P=0.008).结论 HCMV感染与IL-12亚基p40 3’非翻译区+1188(A/C)位点基因多态性具有相关性,其中C等位基因可能是HCMV活动性感染的遗传易感基因,携带C等位基因的个体可能更利于病毒的复制.

     

    Abstract: Objective To investigate the status of single nucleotide polymorphism(SNP) of interleukin-12p40(IL-12p40) 3' untranslated region(3' UTR)+1188(A/C) site and cytomegalovirus infection in infants,and to find the association of IL-12p40 genetic polymorphism with human cytomegalovirus(HCMV) infection.Methods Infants with activa HCMV infection(n=125),infants with latent HCMV infection(n=108),and healthy controls(n=110) were genotyped and analyzed for the SNP of IL-12p40+1188(A/C) using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) and DNA sequencing.HCMV DNA in urine of the infants with active HCMV infection were analyzed using fluorescence quantity-polymerase chain reaction(FQ-PCR).Results The frequency distribution of AA,AC and CC genotype of IL-12p40+1188(A/C) site in the infants with active and latent HCMV infection and healthy controls existed a significant difference(χ2=10.542,P=0.032).Frequencies of A and C allele of IL-12p40+1188(A/C) site were 52.8% and 47.2% in the infants with active HCMV infection,63.4% and 36.6% in the infants with latent HCMV infection,and 67.7% and 32.3% in the control group,with significant differences among the three grouops(χ2=11.777,P=0.003).There were significant differences in the allele frequency distribution between the infants with active HCMV infection(χ2=10.930,P=0.001; odds ratioORA=0.805,95% confidence intervalCI:0.703-0.922; ORC=1.372,95% CI:1.141-1.650).There were significant differences of HCMV DNA in urine in AA,AC and CC genotype of the infants with HCMV active infection(F=9.325,P=0.006).Conclusion There is a relationship between SNP of IL-12p40+1188(A/C) site and the susceptibility to HCMV infection,and the carriers with C allele may be susceptible to develop active HCMV infection.
