江苏省单纯疱疹病毒2型分子流行病学分析

Molecular epidemiologic analysis of HSV-2 in Jiangsu province

  • 摘要: 目的 了解江苏省性病门诊就诊者中单纯疱疹病毒(HSV-1、HSV-2)感染流行状况和主要流行病毒株进化特征。方法 选取江苏省苏南、苏中、苏北3个地区16家性病门诊就诊者116例,采集生殖器疱液标本,采用聚合酶链反应(PCR)扩增HSV-1和HSV-2 TK基因,对阳性样品进行序列分析,与国际参考株进行对比,构建进化树。结果 116例性病就诊者中,HSV-1阳性4例,感染率为3.5%;HSV-2阳性36例,感染率为31.0%;PCR扩增到1130 bp大小的目的基因片断;进化树分析结果显示,苏南地区HSV-2流行毒株全部聚集形成1个病毒簇;苏中地区流行株除1株外,其他也聚集成簇;苏北流行株差异较大,形成>4个独立流行簇,其中2株在核酸序列上与苏南株更接近。结论 HSV-2是江苏省生殖器疱疹主要病原体,HSV-2流行株具有明显的地域特点,并已形成不同地区间传播。

     

    Abstract: Objective To assess the prevalence of herpes simplex virus-1 (HSV-1) and herpes simplex virus-2 (HSV-2) among patients with sexual transmission disease and the phylogenetic characteristic of predominant HSV-2.Methods Totally 116 samples were collected from 16 outpatients from different areas in Jiangsu province.The TK gene of HSV-1 and HSV-2 were amplified,and the positive products were sequenced.After alignment with reference strain NC001798,the phylogenetic tree was constructed.Results Among 116 outpatients with sexually transmitted diseases,4 were HSV-1 positive with a prevalence of 3.5% and 36 were HSV-2 positive,with a prevalence of 31.0%.The virus sequences of HSV-2 from Su'nan region clustered together.The sequences of HSV-2 from Suzhong region also clustered except for SZ8.Conclusion HSV-2 was the major pathogen of genital herpes in Jiangsu province and the prevalence of HSV-2 was obviously different among various areas.
