黄芪甲苷对免疫性肝损伤大鼠保护作用

Protective effect of astragaloside Ⅳ on immunological liver injury in rats

  • 摘要: 目的 探讨黄芪甲苷对卡介苗(BCG)和脂多糖(LPS)诱导肝损伤的保护作用。方法 60只SD大鼠随机分为对照组,模型组(BCG+LPS),黄芪甲苷(20、40、80 mg/kg)组和联苯双酯(150 mg/kg)组,采用BCG 2.5 mg/只静脉注射致敏大鼠,第10 d给予 LPS 75 μg/只静脉攻击,复制大鼠免疫性肝损伤模型,黄芪甲苷或联苯双酯组分别同时灌胃给予黄芪甲苷或联苯双酯,连续10 d,分光光度法测定大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST),超氧化物歧化酶(SOD),丙二醛(MDA)含量;观察肝组织形态学变化;称重法测定肝脏、脾脏指数;酶联免疫吸附法测定血清中TNF-α,IL-6水平。结果 与模型组比较,40、80 mg/kg黄芪甲苷组大鼠血清AST、ALT 活性降低、肝脏、脾脏指数降低;与对照组比较,模型组大鼠血清中TNF-α(60±4.2) pg/mL、IL-6(289±12. 8) pg/mL水平、MDA含量(25.3±2.8) nmol/mg prot升高,SOD活性(72.2±9.8) U/mg prot下降;与模型组比较,80 mg/kg黄芪甲苷组大鼠血清中TNF-α(26±1.5) pg/mL、IL-6(139±13.8) pg/mL水平、MDA含量(14.2±2.9) nmol/mg prot明显下降,SOD活性(108.1±8.3) U/mg prot升高。结论 黄芪甲苷对大鼠免疫性肝损伤具有一定保护作用。

     

    Abstract: Objective To study protective effect of astragaloside Ⅳ on immunological liver injury induced by bacillus Calmette-Guerin(BCG) and lipopolysaccharide(LPS).Methods Sixty Sprague-Dawley(SD) rats were randomly divided into 6 groups:normal group, model group, different dose of astragaloside Ⅳ groups(20, 40, 80 mg/kg), and biphenyl positive control group(150 mg/kg).The model of rat immunological liver injury was induced by injecting 2.5 mg/rat BCG and 75 μg/rat LPS at the tenth day.The content of alanine transaminase(ALT), aspirate aminotransferase(AST), superoxide dismutase(SOD), and malondialdehyde(MDA) were measured by spectrophotometric method.Liver pathological changes were observed with hematoxylin-eosin(HE) staining.The liver index and spleen index were determined by weighting method.The content of tumor necrosis factor α(TNF-α) and interleukin-6(IL-6) were determined with enzyme-linked immunosorbent assay(ELISA).Results Compared with the model group, the activity of ALT, AST and the liver and spleen index were decreased in astragaloside Ⅳ(40, 80 mg/kg) groups.Compared with the control group, the content of TNF-α(60±4.2 pg/mL), IL-6(289±12.8 pg/mL)and MDA(25.3±2.8 nmol/mg prot)increased and the activity of SOD(72.2±9.8 U/mg prot)decreased in model group.Compared with the model group, the content of TNF-α(26±1.5 pg/mL), IL-6(139±13.8 pg/mL)and MDA(14.2±2.9 nmol/mg prot)decreased significantly, and the SOD activity(108.1±8.3 U/mg prot)increased significantly in astragaloside Ⅳ(80 mg/kg) group.Conclusion Astragaloside Ⅳ has a protective effect on immunological liver injury.
