Interaction of polymorphisms of ameloblastin and environmental factors in endemic fluorosis

  • 摘要: 目的 探讨成釉细胞蛋白(AMBN)基因多态性与环境因素在燃煤型地方性氟中毒中的交互作用。方法 采用1:1病例对照研究方法,选取重庆市巫山县笃坪乡和奉节县兴隆镇2个氟病区人群作为观察对象:病例组121例、病区对照121人,并利用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)法对AMBN基因多态性进行分析。应用条件logistic回归模型分析重庆市人群中氟中毒发生的环境及遗传因素,根据交互作用指标判断该基因与环境的交互作用。结果 抽样地区人群的尿氟水平(F)≥0.95 mg/L及AMBN基因的7号外显子(538_540delGGA)的-/-(GGA完全缺失)基因型与13号外显子(986C>T)的CT基因型是该人群氟中毒的危险因素,两者分别存在交互作用(OR=5.600,γ=1.543;OR=4.375,γ=1.296)。结论 携带(538_540delGGA)-/-基因型、(986C>T)CT基因型与尿氟含量(F≥0.95 mg/L)在燃煤型氟中毒中存在一定交互作用,燃煤型氟中毒与遗传及环境因素交互作用有关。

     

    Abstract: Objective To explore the interaction of polymorphisms of ameloblastin(AMBN) gene and environmental factors in endemic fluorosis.Methods A case-control study was carried out and Duping township of Wushan county and Xinglong township of Fengjie county in Chongqing municipality were chosen as fluoride areas.The observation subjects were divided into a case group(121) and a control group(121 in the endemic areas).Polymerase chain-reaction restriction fragment length polymorphism(PCR-RFLP) was used to determine the polymorphisms of AMBN.Interaction indexes(γ)were calculated to determine the type of gene-environmental interation.Conditioned logistic regression analysis was adopted in data analysis with SPSS.Results Urinary fluoride of ≥0.95 mg/L was interacted with the genotypes(538_540delGGA)-/- of exon 7(odds ratioOR=5.600, y=1.543)and genotype(986C>T)CT of exon 13(OR=4.375, y=1.296) in AMBN in the incidence of endemic fluorosis.Conclusion Genotypes of 538-540 delGGA-/- and(986C>T)CT in AMBN may interact with urinary fluoride(≥0.95 mg/L)in the incidence of coal-burning fluorosis.
