天然植物蜈蚣草对大鼠排砷效果评价

Impact of natural plant Pteris vittata on arsenic excretion in rats

  • 摘要: 目的 观察天然植物蜈蚣草对大鼠的排砷效果。方法 大鼠50只,随机分成对照组、模型组、阳性对照组及蜈蚣草低、高剂量组,每组10只;除对照组外,各组动物每天腹腔注射亚砷酸钠溶液(2 mg/kg),每周连续注射5 d,共4周;蜈蚣草组连续6周给予含蜈蚣草饲料喂养,阳性对照组肌肉注射二巯基丙磺酸钠(DMPS),动态检测大鼠尿砷、血砷含量;停砷2周时取动物血测定血红蛋白、肌酐、尿素氮和天冬氨酸氨基转移酶(ALT),剖检动物并计算脏器系数、骨髓嗜多染红细胞微核发生率。结果 在染砷4周时,蜈蚣草高、低剂量组、模型组及DMPS组大鼠的尿砷含量分别为(6.06±0.61)、(3.45±0.79)、(2.48±0.27)、(6.63±0.68) mg/L,与模型组比较,高剂量蜈蚣草组大鼠尿砷含量明显升高,差异有统计学意义(P<0.01),其排砷作用与DMPS相近;各组大鼠血红蛋白、肌酐、尿素氮、ALT及肝、肾等脏器系数无明显改变;高剂量蜈蚣草对砷所致的大鼠骨髓嗜多染红细胞微核发生率增高具有明显抑制作用。结论 蜈蚣草制剂具有一定的排砷效果,42 d 连续喂养未发现其对大鼠机体有明显损害作用。

     

    Abstract: Objective To observe the impact of natural plant Pteris vittata on arsenic(As)excretion in rats.Methods Fifty rats were randomly divited into normal,model,positive control,low and high dose group(10 rats in each group).Excepted for the normol group,all groups received intraperitoneal injection of sodium arsenite(2 mg/kg)5 days a week for 4 weeks.Low and high dose group received feeds containing Pteris vittata for 6 weeks.Positive control group received intramuscular injection of sodium dimercapto-sulphonate(DMPS).Urine and blood As in rats was detected dynamically.Two weeks after the As injection,blood heamoglobin(Hb),creatinine(Cr),blood urea nitrogen(BUN),and aspartate transaminase(ALT)were detected.Then,all the rats were dissected and their organ coefficient and micronucleus rate of polychromatic erythrocytes of bone marrow were calculated and compared.Results In the 4th weeks of As exposure,urine As in high dose,low dose,model and DMPS group were 6.06±0.61,3.45±0.79,2.48±0.27,and 6.63±0.68 mg/L,respectively.Urine As in high dose group was significantly higher than that of the model group(P<0.01).The As excretion of high dose group was similar to that of DMPS group.There were no obvious changes observed in Hb,Cr,BUN,ALT,and organ coefficient of liver and kidney.The increased micronucleus rate of polychromatic erythrocytes induced by As was inhibited in high dose group.Conclusion Extracted Pteris vittata has obvious impact on As excretion.No obvious damage effect was observed in the rats with continuous feeding of Pteris vittata for 42 days.
