芪葛山菜胶囊对高血糖模型小鼠血糖影响

Effect of Qige-Shancai compound capsule on mice with high blood glucose

  • 摘要: 目的探讨芪葛山菜胶囊对高血糖模型小鼠血糖的影响。方法SPF级昆明种成年小鼠60只, 随机分为6组, 每组10只, 溶剂对照组、芪葛山菜对照组、模型组、芪葛山菜低、中、高剂量组(分别灌胃给予0.45、0.90、1.35 g/kg, 连续30 d);模型组和芪葛山菜低、中、高剂量组小鼠尾静脉注射四氧嘧啶建立高血糖动物模型, 观察小鼠一般生长状态及体重变化, 采用血糖仪检测各组小鼠空腹血糖值、采用糖耐量实验比较各组小鼠血糖曲线下面积变化。结果与溶剂对照组比较, 模型组、芪葛山菜低、中、高剂量组、芪葛山菜对照组小鼠体重、体重增重均无明显变化;与溶剂对照组(5.57±1.19)mmol/L比较, 模型组小鼠空腹血糖(21.15±1.76)mmol/L明显升高, 差异有统计学意义(P<0.01);与模型组比较, 芪葛山菜0.90、1.35 g/kg剂量组小鼠空腹血糖值(15.96±1.91)、(15.43±2.19)mmol/L明显下降, 差异有统计学意义(P<0.01), 血糖下降百分率分别为(19.97±12.79)%、(24.22±16.47)%;与模型组(57.22±3.83)比较, 芪葛山菜1.35 g/kg剂量组小鼠血糖曲线下面积(52.70±5.44)明显下降, 差异有统计学意义(P<0.05);1.35 g/kg芪葛山菜对血糖正常小鼠的空腹血糖无明显影响。结论芪葛山菜胶囊可明显降低四氧嘧啶所致高血糖小鼠的空腹血糖水平, 且对小鼠一般生长发育无明显影响。

     

    Abstract: ObjectiveTo investigate the effect of Qige-Shancai compound capsule(QSCC,a mixture of astragalus,kudzu root,Gynura divaricata,and wolfberry fruit with total saponins of 350 mg/100 g and puerarin of 1.5 g/100 g) on mice with high blood glucose.MethodsSixty special pathogen free(SPF) adult Kunming mice were randomly divided into 6 groups(10 mice in each group):solvent control,QSCC control,model,and low-,medium-,high-dose groups administered with QSCC of 0.45,0.90,1.35 g/kg by gavage once a day for 30 days.High blood glucose of the mice were induced by injecting alloxan through caudal vein.General growth status and weight change in the mice were observed ; fasting blood glucose was measured with glucometer; and changes in the area under the curve of blood glucose determined with glucose tolerance test were compared among the groups.ResultsBody weight and weight gain showed no significant differences between the solvent control and the other groups.Compared with that of the control group,fasting blood glucose significantly increased in the model group(21.15±1.76 vs.5.57±1.19 mmol/L)(P<0.01).Compared with the model group,fasting blood glucose of the mice in 0.90 and 1.35 g/kg QGCC groups decreased to 15.96±1.91 and 15.43±2.19 mmol/L significantly(both P<0.01) with the decrease rates of 19.97±12.79% and 24.22±16.47%.A significant decline in the area under blood glucose curve was observed in the mice treated with 1.35 g/kg QGCC compared with the model group(52.70±5.44 vs.57.22±3.83)(P<0.01),but no significant effect of 1.35 g/kg QGCC on fasting blood glucose was observed in normal mice.ConclusionQGCC could reduce fasting blood glucose of mice with high blood glucose and has no significant impact on general growth in mice.
