细粒棘球绦虫Eg18抗原表位预测

Prediction of Eg18 antigen epitopes of Echinococcus granulosus

  • 摘要: 目的 通过生物信息学技术预测细粒棘球绦虫Eg18 抗原B、T细胞表位,为细粒棘球绦虫筛选具有保护性抗原表位的抗原提供理论基础。方法 使用Expasy系统预测理化性质;二、三级结构的预测使用在线预测网站Predict Protein;通过在线软件ABCpred、LEPS结合二级结构、亲水性指数、表面可及性指数、柔韧性指数对B细胞表位进行预测;应用SYFPEITHI及IEDB软件预测MHC-Ⅰ类HLA-A0201限制性T细胞表位。结果 Eg18抗原二级结构中α-螺旋结构占氨基酸残基总数的98.1%,无规则卷曲占1.9%,无β-折叠结构;选出具有优势的30~41、55~71、110~126、136~144四个可能为Eg18蛋白B细胞表位的氨基酸序列区域;17~27、32~41、61~72、96~105四个较有可能形成T抗原表位的氨基酸序列区域。结论 通过生物信息学预测Eg18 抗原可能有4个B细胞表位且为构象表位,有4个T抗原表位,可为进一步了解Eg18 抗原提供参考。

     

    Abstract: Objective To predict and analyze B cell and T cell epitopes of Echinococcus granulosus Eg18,and to provide basic information for screening protective antigens of Echinococcus granulosus.Methods The physicochemical property of Eg18 antigen was predicted using Expasy system and the secondary and tertiary structure of Eg18 antigen were predicted using online software Predict Protein.The secondary structure,hydrophilic,surface accessibility index,and flexibility index of B cell epitopes were predicted with online software ABCpred and LEPS;the restrictive T cell epitopes of major histocompatibility complex-I (MHC-Ⅰ) type HLA-A0201was predicted using SYFPEITHI and IEDB software.Results The ratio of total amino acid residues involved in alpha helix of Eg18 antigen secondary structure are 98.1% and that of in random coil are 1.9%.No β-folding was observed in the special structure of the antigen.Four ranges of animo acid sequence (30-41,55-71,110-1261,and 36-144) in Eg18 antigen were recognized as the possible B cell antigen epitopes of Eg18 and another four ranges (17-27,32-41,61-72,and 96-105) might be the possible T cell antigen epitopes of Eg18.Conclusion Four B cell epitopes and four T cell epitopes of Echinococcus granulosus Eg18 were predicted using bioinformatics technology,which provide a reference for further research on the Eg18 antigen.
