天然食物小根蒜诱导S180细胞凋亡作用的研究

摘要:目的 探讨天然食物小根蒜体外诱导小鼠S180肿瘤细胞凋亡作用, 寻找开发预防和治疗肿瘤的新资源及提取有效成分应用于临床。方法 以不同浓度的小根蒜水提取液和小鼠S180肿瘤细胞共同体外培养12和24h, 采用流式细胞仪碘化丙啶(PI)染色法, 测定细胞凋亡百分率及对细胞周期的影响。结果 小根蒜水提取液可诱导小鼠S180肿瘤细胞凋亡, 其凋亡百分率(AP%)随着浓度的增加及作用时间的延长而增加。作用24h, 500mg/ml小根蒜AP%为43.28%.对细胞周期的影响, 表现在S期、G2/M期细胞百分率下降。结论 天然食物小根蒜能诱导小鼠S180肿瘤细胞凋亡, 并抑制DNA合成及细胞增殖。Abstract:Objective To study the effect of AMB on mouse S180 tumor cell apoptosis in vitro.Methods S180 tumor cells were treated with the water extract of AMB in different concentrations and cultivated in vitro for 12h and 24h.Results The apoptosis rate of mouse S180 tuor cells treated with AMB at the concentration of 500mg/ml for 24 h was 43.28%.The apoptosis-inducing effect of AMB was dose-dependent and reaction time dependent.Conclusion AMB can kill mouse S180 tumor cells by inducing cell apoptosis and inhibit S180 tumor cells DNA synthesize.
